Effect of miR-15a-5p on the proliferation of gastric cancer cells

目的观察miR-15a-5p对胃癌细胞增殖的影响。方法将miR-15a-5p mimics、mimics control、miR-15a-5p inhibitor、inhibitor control分别转染至胃癌SGC-7901和BGC-823细胞,应用实时定量PCR分别检测miR-15a-5p的表达水平以验证miR-15a-5p mimics对miR-15a-5p表达的影响及miR-15a-5p inhibitor对miR-15a-5p表达的影响。CCK-8实验检测miR-15a-5p mimics及miR-15a-5p inhibitor对胃癌SGC-7901和BGC-823细胞增殖的影响。结果实时定量PCR表明与转染mimics control相比,转染miR-15a-5p mimics后SGC-7901细胞和BGC-823细胞中miR-15a-5p的表达水平显著升高(P <0. 05)。与转染inhibitor control相比,转染miR-15a-5p inhibitor后SGC-7901和BGC-823细胞中miR-15a-5p的表达有下降趋势,但差异无统计学意义(P> 0. 05)。CCK-8实验表明过表达miR-15a-5p可抑制胃癌细胞SGC-7901和BGC-823的增殖(P <0. 05),miR-15a-5p inhibitor促进SGC-7901和BGC-823细胞增殖(P <0. 05)。结论 miR-15a-5p可能抑制胃癌细胞的增殖。

Objective To investigate the effect of miR-15 a-5 p on the proliferation of gastric cancer cells. Methods The miR-15 a-5 p mimics,mimics control,miR-15 a-5 p inhibitor or inhibitor control were transfected into SGC-7901 and BGC-823 cells,respectively.Real-time quantitative PCR was used to verify the expression level of miR-15 a-5 p. CCK-8 assay was performed to detect the effect of miR-15 a-5 p mimics and miR-15 a-5 p inhibitors on the proliferation of SGC-7901 and BGC-823 cells. Results Real-time quantitative PCR confirmed that the expression of miR-15 a-5 p was significantly increased in SGC-7901 cells and BGC-823 cells after transfection with miR-15 a-5 p mimics compared with mimics control( P < 0. 05). The expression of miR-15 a-5 p was decreased in SGC-7901 and BGC-823 cells after transfection with miR-15 a-5 p inhibitor compared with inhibitor control,however,the difference was not statistically significant( P > 0. 05). CCK-8 results showed that overexpression of miR-15 a-5 p suppressed the proliferation of SGC-7901 and BGC-823 cells( P < 0. 05),while miR-15 a-5 p inhibitor promoted the proliferation of SGC-7901 and BGC-823 cells( P < 0. 05). Conclusion The miR-15 a-5 p could suppress the proliferation of gastric cancer cells.
