摘要
目的建立银杏叶标准汤的质量评价标准及指纹图谱。方法采用高效液相色谱(HPLC)法测定银杏标准汤的待测成分含量,色谱柱为Welchrom C18柱(250 mm×4. 6 mm,5μm),流动相为甲醇-0. 4%磷酸溶液(50∶50,V/V),流速为1. 0 m L/min,检测波长为360 nm,柱温为30℃,进样量为10μL;计算出膏率、转移率;以山柰素峰为参照峰,采用中药色谱指纹图谱相似度评价系统(2004A版)检测,并进行相似度比较。结果标准汤出膏率为(13. 89±1. 42)%;总黄酮醇苷转移率为(55. 93±13. 79)%;建立了15批标准汤的特征图谱,指纹图谱批次间相似度均大于0. 95。结论该研究中所建立的指纹图谱和定量分析方法准确、稳定、重复性好,可用于评价银杏叶标准汤的质量。
Objective To establish the quality evaluation standard and fingerprint of Ginkgo Folium standard decoction. Methods The content of the components in Ginkgo Folium standard decoction was determined by HPLC,the chromatographic column was Welchrom C18 column( 250 mm × 4. 6 mm,5 μm),the mobile phase was methanol-0. 4% phosphoric acid solution( 50 ∶ 50,V/V),the flow rate was 1. 0 m L/min,the detection wavelength was 360 nm,the column temperature was 30 ℃,and the sample size was 10 μL. The extraction rate and transfer rate were calculated,the kaempferol peak was used as the reference peak,and the similarity was detected and compared by the Similarity Evaluation System for Chromatographic Fingerprints of TCM( Version 2004). Results The extraction rate of the standard decoction was( 13. 89 ± 1. 42)%. The transfer rate of total flavonol glycosides was( 55. 93 ± 13. 79)%. The characteristic chromatograms of 15 batches of standard decoction were established,and the similarity among batches of fingerprint chromatograms was more than 0. 95. Conclusion The fingerprint and quantitative analysis methods established in this study are accurate,stable and reproducible,which can be used to evaluate the quality of Ginkgo Folium standard decoction.
