石榴皮多酚的抗氧化性及其对PC12细胞增殖的影响

Antioxidation of pomegranate peel polyphenol and its effect on PC12 cell proliferation

目的研究石榴皮多酚提取物的体外抗氧化能力和对PC12细胞增殖的影响。方法用溶剂提取法从石榴皮中提取多酚类物质,采用DPPH和ABTS法测定石榴皮多酚提取物清除自由基的能力,用MTT法研究石榴皮多酚提取物对细胞增殖的影响。结果石榴皮提取物中的总多酚含量为其干重的18.40%,石榴皮多酚提取液清除DPPH·自由基半抑制浓度(IC_(50))为3.64μg/mL,清除ABTS^+自由基半抑制浓度(IC_(50))为2.63μg/mL。结论石榴皮多酚提取物具有较强的清除自由基能力,可以作为天然抗氧化剂清除体内正常代谢所产生的自由基,同时发现石榴皮多酚可在体外诱导PC12凋亡,具有较好的抗肿瘤活性,为石榴皮的综合应用提供理论基础。

Objective: To study the antioxidant capacity in vitro of pomegranate peel polyphenol extract and its effect on PC12 cell proliferation.Methods: Pomegranate peel polyphenols was extracted by solvent extraction method,the ability of scavenging free radicals was determine by DPPH and ABTS method,and pomegranate peel polyphenols extract effects on cell proliferation was the determined by MTT method.Results: Pomegranate peel extract was 18.40% of the total polyphenol content of dry weight,pomegranate peel polyphenols extract to remove DPPH radical half inhibitory concentration (IC 50 ) was 3.64μg/mL, and remove ABTS free radicals half inhibitory concentration (IC 50 ) was 2.63μg/mL.Conclusion: Pomegranate peel polyphenols extract has the strong ability of scavenging free radicals,which can be used as natural antioxidants in the body by scavenging free radicals produced by normal metabolism.And it is also found that pomegranate peel polyphenols can be induced PC12 apoptosis in vitro and have good antitumor activity,providing theoretical basis for pomegranate peel of comprehensive application.
