摘要
为提高乳酸片球菌ZPA017的γ-氨基丁酸(GABA)产量,本试验通过单因素筛选和响应面法,对其发酵培养基和培养条件进行了优化。结果表明:乳酸片球菌ZPA017产GABA的最佳培养基配方为:D-果糖18g/L、大豆蛋白胨40g/L、乙酸钠3g/L、磷酸氢二钾1.37g/L、柠檬酸氢二铵2g/L、硫酸镁0.63g/L、硫酸锰0.29g/L、吐温-801mL/L。在此基础上,采用单因素筛选的方法,优化得到乳酸片球菌ZPA017产GABA的最佳发酵条件为:温度37℃、初始pH6.5、接种比例1%、发酵时间30h。乳酸片球菌ZPA017在最佳培养基和发酵条件下培养,发酵液中γ-氨基丁酸的含量达1.036g/L,是优化前产量的3.77倍。
To improve the production ofγ-aminobutyric acid(GABA)by Pediococcus acidilactici ZPA017,the experiment used single factor analysis and orthogonal design experiment to optimize the cultivation medium components and fermentation conditions.The results showed that the optimum medium formulation for GABA by Pediococcus acidilactici ZPA017 were D-fructose 18 g/L,soy peptone 40 g/L,sodium acetate 3 g/L,K2HPO4 1.37 g/L,diammonium hydrogen citrate 2 g/L,MgSO4 0.63 g/L,MnSO4 0.29 g/L,tween-80 1 mL/L.The optimum cultivation conditions were as follows:incubation temperature was 37℃,initiation pH was 6.5,inoculation volume was 1%and the fermentation time was 30 h.The content of GABA was up to 1.036 g/L after the optimization,which was about 3.77 times higher prior to previous optimization.
