摘要
目的研究中年与老年猕猴单个核细胞中miRNA表达差异,并预测其调控的靶基因。方法利用基于二代测序的miRNA测序方法,对中年和老年猕猴外周血中单个核细胞的miRNA进行测序,利用生物信息学方法对测序数据进行统计分析。结果中年与老年猕猴外周血单个核细胞的差异表达miRNA有55个,其中21个为已知miRNA,34个为新发现miRNA。已知miRNA中,有4个miRNA为上调表达,17个miRNA为下调表达。通过生物信息学分析预测发现,已知差异miRNA的调控靶基因1634个,分别富集在35个KEGG信号通路上。结论发现中年和老年猕猴血单个核细胞差异表达的miRNA,并预测出其调控的靶基因。
Objective To study the differential expression of miRNA in mononuclear cells of middle-aged and old-aged rhesus macaques and predict the target genes regulated by miRNA. Methods The miRNA sequencing technology based on next-generation sequencing was applied to miRNA in peripheral blood mononuclear cells(PBMCs) of middle-aged and old-aged rhesus macaques, and the sequencing data were statistically analyzed with bioinformatics methods. Results There were 55 miRNAs with differential expression in PBMCs of middle-aged and old-aged rhesus macaques, including 21 known miRNAs and 34 newly discovered miRNAs.Among the known miRNAs, four were up-regulated, and 17 were down-regulated. The bioinformatics analysis and prediction showed that there were 1634 target genes regulated by the known miRNA with differential expression, and such target genes were enriched in35 KEGG signaling pathways respectively. Conclusion The miRNAs with differential expression in PBMCs of middle-aged and old-aged rhesus macaques have been found and the target genes regulated by miRNAs have been predicted.
