深度测序解析年龄及UCMSC治疗对猕猴lncRNA的影响

Deep sequencing analysis of the effects of age and UCMSC treatment on lncRNA in rhesus macaques

摘要 目的分析年龄和脐带间充质干细胞(UCMSC)治疗对外周血单个核细胞长链非编码RNA(lncRNA)的影响。方法分别采集、分离幼年、中年、老年和老年经UCMSC治疗后的4种不同生理状态的猕猴外周血单个核细胞,提取全基因组RNA,进行转录组测序,通过生物信息学比较筛选出差异lncRNA并进行分组比较。结果幼年与老年、中年相比,单个核细胞表达上调的lncRNA分别为54和52个,表达下调的lncRNA分别为49和46个;中年与老年相比,表达上调和下调的lncRNA分别为48和63个;老年与经UCMSC治疗组相比,表达上调和下调的lncRNA分别为41和68个。聚类分析发现,随年龄增长,幼年高表达lncRNA减少,而低表达lncRNA增加;UCMSC治疗可使低表达lncRNA升高,而高表达lncRNA降低。结论lncRNA表达谱随年龄的增加而改变,UCMSC治疗可使衰老状态的lncRNA表达谱趋向年轻化方向。 Objective To analyze the effects of age and umbilical cord mesenchymal stem cell(UCMSC) treatment on the long noncoding RNA(lncRNA) in peripheral blood mononuclear cells(PBMCs). Methods The PBMCs of rhesus macaques at different physiological states from a young-age group, a middle-age group, an old-age group and an old-age group receiving UCMSC treatment were collected and separated respectively, and the whole-genome RNA was extracted for transcriptome sequencing. By comparison of bioinformatics, the differential lncRNAs were screened out and compared among these groups. Results When compared between the young-age group and the old-age group, the number of lncRNA with up-regulated expression and down-regulated expression in mononuclear cells was 54 and 52 respectively. When compared between the young-age group and the middle-age group, the number of lncRNA with up-regulated expression and down-regulated expression in mononuclear cells was 49 and 46 respectively. When compared between the middle-age group and the old-age group, the number of lncRNA with up-regulated expression and downregulated expression in mononuclear cells was 48 and 63 respectively. When compared between the old-age group and the old-age group receiving UCMSC treatment, the number of lncRNA with up-regulated expression and down-regulated expression in mononuclear cells was 41 and 68 respectively. Cluster analysis indicated that the number of lncRNA with high expression in young rhesus macaques decreased with age, while the number of lncRNA with low expression increased with age. UCMSC treatment resulted in the rise of lncRNA with low expression and the decline of lncRNA with high expression. Conclusion The expression profile of lncRNA changes with age, and the expression of lncRNA in aging status can be up-regulated with UCMSC treatment.
