摘要 目的探讨Cx46与晶状体上皮细胞凋亡和年龄相关性白内障的相关性。方法建立H2O2(浓度为100μM)介导的晶状体上皮细胞(SRA01/04)凋亡模型,用Western blotting实验检测Cx46蛋白表达量。利用MTT技术分析细胞生存率。利用Hoechst33342染料固定分析细胞凋亡情况。结果试验数据阐明H2O2(浓度为100μM)是导致晶状体上皮细胞凋亡的重要诱导因素。Cx46低表达与H2O2介导的人晶状体上皮细胞凋亡相关[蛋白表达量:H2O250μM(177.67±3.65)vsH2O2100μM(139.02±2.13)(P<0.05)]。Cx46过表达能抑制H2O2介导的人晶状体上皮细胞凋亡进程[凋亡率:Cx46(24.36±3.13)%vsH2O2(43.65±7.51)%(P<0.01)]。结论研究数据确定凋亡的晶状体上皮细胞中Cx46低表达与年龄相关性白内障的发生直接相关。 Objective To explore the correlation between Cx46 and lens epithelial cell apoptosis,age-related cataract.Methods The apoptosis model of lens epithelial cells(SRA01/04)mediated by H2O2(concentration of 100μM)was established.The expression of Cx46 protein was detected by Western blotting.Cell viability was analyzed using MTT technology.Cell apoptosis was analyzed by Hoechst 33342 dye fixation.Results The experimental data demonstrated that H2O2(concentration of 100μM)was an important inducer of apoptosis in lens epithelial cells.Low expression of Cx46 was associated with H2O2-mediated apoptosis of human lens epithelial cells[protein expression:H2O2 50μM(177.67±3.65)vs H2O2 100μM(139.02±2.13)](P<0.05).Overexpression of Cx46 inhibited H2O2-mediated apoptosis in human lens epithelial cells[apoptosis rate:Cx46(24.36±3.13)%vs H2O2(43.65±7.51)%](P<0.01).Conclusion The study data determine that low expression of Cx46 in apoptotic lens epithelial cells is directly associated with the development of age-related cataract.
