兴安杜鹃基本培养基和外殖体选择的初步研究

Preliminary Study on the Selection of Basic Culture Medium and Explants of Rhododendron dauricum

[目的]研究兴安杜鹃苗木快繁技术,为兴安杜鹃大规模园林绿化应用提供技术保障。[方法]以兴安杜鹃的叶芽、花芽、叶片、茎尖、茎段为外植体,以MS、WPM、Read、B5为培养基,采用不同采集时间、不同消毒方法,通过接种、培养、观察和对比分析,对兴安杜鹃的快速繁殖进行研究。[结果]兴安杜鹃组织培养最佳基本培养基为WPM培养基;最佳外植体为1.5~2.0cm茎尖和茎段;最佳灭菌方法为70%乙醇30s结合0.1%HgCl210min;最佳取材时间为3月中旬至5月中旬。[结论]该研究可为绿化苗木生产、城镇绿化及乡村振兴生态宜居目标的实现提供支撑。

[Objective]To study the rapid propagation technology of Rhododendron dauricum seedlings, and provide technical support for the large-scale landscaping application of Rhododendron dauricum.[Method]The leaf buds, flower buds, leaves, stem tips and stem segments of Xing'an Rhododendron were used as explants, and MS, WPM, Read B5 were used as the medium.The rapid propagation of Rhododendron dauricum studied by using explants with different sampling time and disinfection methods through inoculation, culture, observation and comparative analysis.[Result]The best basic medium for tissue culture of Rhododendron dauricum was WPM;the optimal explants was1.5-2.0 cm long stem tips and stem segments;the best sterilization method on the surface of explants was 70% alcohol 30 s and then combined with 0.1% HgCl210 min;the best season for sampling time was from mid-March to mid-May.[Conclusion]This research can provide technique to reproduction of greening seedlings,urban greening and realization of ecological livable target in rural vitalization.
