摘要 实验旨在研究 BMAL1基因对睾丸间质细胞凋亡的影响。实验以小鼠睾丸间质细胞系为研究对象,使用小干扰 RNA(siRNA)抑制 BMAL1的表达,使用流式细胞术、实时荧光定量 PCR(qRT-PCR)和蛋白印迹(Western Blot)检测细胞凋亡水平。结果表明:干扰组睾丸间质细胞凋亡水平显著上升(P<0.05),且伴随着促凋亡蛋白 BAX 显著上调和抑凋亡蛋白 BCL-2 表达下调(P<0.05)。结果显示,BMAL1作为核心的生物钟基因,可抑制小鼠睾丸间质细胞的凋亡。 The aim of the study was to investigate the effect of BMAL1 gene on apoptosis of Leydig cells.. In this study, mouse Leydig cell line was used as the research object, and small interfering RNA (siRNA) was used to inhibit the expression of BMAL1. Using flow cytometry, real-time quantitative PCR (qRT-PCR) and Western Blot to test the level of apoptosis. These results suggested that the apoptosis level of Leydig cells in the interfering group was signi ficantly increased, and the pro-apoptotic protein BAX was signi ficantly up-regulated and the anti-apoptotic protein BCL-2 was down-regulated. These results demonstrated that BMAL1, as the core clock gene, inhibited the apoptosis of mouse Leydig cells.
