基于特征图谱分析市售决明子质量现状

Analysis of quality status of Cassia obtusifolia in market by specific chromatogram

目的建立决明子Cassia obtusifolia Linn.HPLC特征图谱,并测定橙黄决明素、大黄酸、大黄素、大黄酚、大黄素甲醚的含有量。方法该药物70%乙醇提取液的分析采用Waters XBridge C18色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-0.05%磷酸,梯度洗脱;体积流量1.0 mL/min;柱温30℃;检测波长285 nm。采用相似度分析、主成分分析对22批样品分析。结果 5种成分在各自范围内线性关系良好(r>0.999 0),加样回收率95.6%~101.3%,RSD 0.45%~3.62%。不同批次样品相似度差别明显,主成分分析结果显示不同来源在化学成分的组分和含有量上存在不同程度的差异。结论该方法稳定可靠,重复性好,可用于市售决明子的质量评价。

AIM To establish the HPLC fingerprints of Cassia obtusifolia Linn. and to determine the contents of aurantio-obtusin, chrysophanic acid, emodin, chrysophanol and physcione. METHODS The analysis of 70% ethanol extract of this drug was performed on a 30 ℃ thermostatic Waters XBridge C18 column, with the mobile phase comprising of acetonitrile-0.05% phosphoric acid flowing at 1.0 mL/min in a gradient manner, and the detection wavelength was set at 285 nm.Twenty-two batches of samples were analyzed by similarity analysis and principal component analysis.RESULTS Five constituents showed good linear relationships within their own ranges(r>0.999 0), whose average recoveries were 95.6%-101.30% with the RSDs of 0.45%-3.62%. The similarity of different batch samples was obviously different, and the principal component analysis results showed that there were different degrees of difference in chemical composition and content among different origins.CONCLUSION This stable, reliable and reproducible method can be used for quality evaluation of C. obtusifolia in market.
