摘要
以甘蓝型绿叶油菜"湘油15号"为母本、野生芥茉型紫叶油菜为父本进行甘芥种间杂交,经8代套袋自交和定向选择获得了1个紫叶性状稳定遗传的重组自交系。利用紫叶亲本和绿叶亲本正交、反交、回交后观测后代性状分离比例,推测紫叶性状受单个显性核基因控制;解剖紫色叶片显微观察紫色分布情况,发现其主要分布于叶片上表皮细胞与临近栅栏组织中;提取并测量紫叶叶片与绿叶叶片中几处花色素含量,发现紫叶叶片花青素含量远远高于绿叶叶片;用不同温度和酸碱条件处理该花青素,发现其在低温和酸性环境下具有很好的稳定性。
A recombinant inbred line with stable inheritance of purple leaf traits is obtained by bagged self-crossing and directional selection for 8 generations. In the experiment, the parents of purple leaf and green leaf are orthogonal, backcrossing and backcrossing, and the separation ratio of the traits in the offspring is observed to infer that the traits of purple leaf are controlled by a single dominant nuclear gene.The purple leaves are dissected and observed microscopically and found to be mainly distributed in the epidermal cells and adjacent palisade tissues.The anthocyanin content in leaves of purple leaves and green leaves is higher than that of green leaves.Handling of the anthocyanins in different temperatures, acid and alkali conditions, it is found that it has very good stability below pH5.0 and under 45℃.
