包葛颗粒剂对小鼠醒酒防醉及急性酒精性肝损伤保护作用的研究 被引量:4

摘要 目的研究包葛颗粒剂对小鼠醒酒防醉及急性酒精性肝损伤的保护作用。方法取昆明小鼠40只,随机分为空白组(蒸馏水0.1 mL/10 g体质量)、酒精肝损伤模型组(蒸馏水0.1 mL/10 g体质量)、包葛颗粒剂低剂量组(包葛颗粒剂2.4 g/kg体质量)、包葛颗粒剂高剂量组(包葛颗粒剂4.8 g/kg体质量)、阳性对照组(水飞蓟宾47 mg/kg体质量),各小组小鼠灌胃给药2 h后,除空白对照组给予蒸馏水外,其他各组小鼠灌胃给予红星二锅头白酒(12 mL/kg体质量),连续灌胃7 d,每天1次,第7天灌胃给酒后观察各组小鼠醉酒潜伏期、醉酒期和睡眠期指标,6 h后小鼠麻醉颈椎脱臼处死,取肝脏,称肝重,计算肝指数,按天冬氨酸氨基转移酶(AST)和丙氨酸氨基转移酶(ALT)试剂盒方法测定肝组织AST,ALT含量,观察HE染色后肝脏病理组织学变化。结果与酒精肝损伤模型组比较,包葛颗粒剂低、高剂量组小鼠醉酒潜伏期延长,睡眠期缩短,醉酒期时间缩短,肝指数、肝组织ALT、AST均降低,差异有统计学意义(P<0.05)。肝脏病理组织学结果显示:与酒精肝损伤模型组比较,包葛颗粒剂低、高剂量组小鼠肝组织损伤减轻,细胞结构相对正常,细胞水肿有一定缓解,脂滴数量明显减少。结论包葛颗粒剂对小鼠具有醒酒防醉作用,对急性酒精性肝损伤可以起到保护作用。 Objective To study the anti-drunk and liver protective effects of Baoge Granules on acute alcoholic liver injury mice. Methods 40 Kunming mice were randomly divided into blank group(distilled water 0.1 mL/10 g body mass), alcoholic liver injury model group(distilled water 0.1 mL/10 g body mass), Baoge Granules low-dose group(Baoge Granules 2.4 g/kg body mass), Baoge Granules high-dose group(Baoge Granules 4.8 g/kg body mass),and positive group(Silibinin 47 mg/kg body mass). They were given distilled water after 2 hours of intragastric administration(except the blank group). All groups were given red star Erguotou Liquor(12 mL/kg body mass) once daily for continuous 7 days,the drunkenness,intoxication and sleepy indexes of the mice in each group were observed. After 6 hours,mice were anesthetized and sacrificed by cervical dislocation. The livers were taken and weighed. The liver index was calculated. Aspartate aminotransferase(AST) and alanine aminotransferase(ALT) levels in liver were determined according to AST and ALT kit method. The histopathological changes of liver were observed after HE staining. Results Compared with the alcoholic liver injury model group,in the low and high-dose groups of Baoge Granules, the drunkness of the mice was significantly prolonged,and the sleep period was significantly shortened,and the drunkness period was shortened. The liver index,liver tissue ALT and AST were greatly reduced, and the difference was statistically significant(P<0.05). The histopathological results of liver showed that compared with the alcoholic liver injury model group,the liver tissue damage of the mice in the low and high dose groups of Baoge Granules was alleviated,and the cell structure was relatively normal,and the cell edema was relieved,and the number of lipid droplets was significantly reduced. Conclusion Baoge Granules have a positive effect of anti-drunk on mice, and can protect against acute alcoholic liver injury.
