7种干果组合对AD模型大鼠行为学及氧化应激水平的影响 被引量:2

摘要 目的分析7种干果组合对AD大鼠行为学及氧化应激水平的影响。方法 65只SD大鼠随机分5组(13只/组),分别为低(10%)、中(15%)、高(20%)浓度干果饲料组、假手术对照组、模型对照组。假手术对照组海马区注射盐水,其余组注射β淀粉样蛋白25-35(Aβ25-35)造模。造模第7天,假手术对照组、模型对照组给予正常饲料,其余组给予含相应浓度干果的饲料,干预40 d。35~40 d进行Morris水迷宫实验。采用酶联免疫吸咐实验(ELISA)试剂盒检测各组大鼠血清、海马匀浆中的丙二醛(MDA)含量以及过氧化氢酶(CAT)和超氧化物歧化酶(SOD)的活性。结果与假手术对照组比较,模型对照组定向航行实验潜伏期、朝向角增加,目标象限进入次数减少,空间探索实验潜伏期时间增加,原平台象限滞留时间百分比、原平台区域穿越次数减少,血清、海马中SOD、CAT含量减少,MDA含量增加,差异有统计学意义(P<0.05)。与模型对照组比较,高浓度组定向航行实验潜伏期、朝向角减少,目标象限进入次数增加,空间探索实验潜伏期时间减少,原平台象限滞留时间百分比、原平台区域穿越次数增加,血清、海马中SOD、CAT含量减少,MDA含量增加,差异有统计学意义(P<0.05)。结论该干果组合可改善AD大鼠的学习、记忆能力,其机制可能与干果组合增强AD大鼠抗氧化能力有关。 Objective To explore the effects of 7 dried fruits′ combinations on Alzheimer′s Disease(AD) rats model. Methods A total of 65 SD rats were randomly divided into 5 groups(with 13 rats in each group) comprised: Sham surgery, model control, low(10%), medium(15%), and high(20%) concentrations. In the sham group, saline was injected into the hippocampus;while in the rest, Aβ25-35 was injected for modeling. On the 7 th day of surgery, the sham operation group and the model group were fed with normal diet, and the other groups were fed with corresponding dried fruit diet continuously for 40 days. On the 35 th to 40 th days, start Morris water maze. The contents of SOD, CAT and MDA in serum and hippocampus were determined by ELISA. Results Compared with the sham operation group, the incubation period and orientation Angle of the model group increased, and the number and time of entering the target quadrant decreased. SOD and CAT in serum and hippocampus decreased and MDA increased, the difference was statistically significant(P<0.05). Compared with the model group, the higher the concentration of dried fruit feed was, the incubation period and orientation Angle were generally in a downward trend, while The Times of entering the target quadrant were on the contrary.CAT content in serum and hippocampus were increased with increasing concentration, MDA content was decreased and SOD content was increased. The SOD, CAT and MDA contents in the serum of the medium concentration group and the serum of the high concentration group and the hippocampus were significantly higher than those of the model group(P<0.05). Conclusion The dried fruit combination was exhibited to be capable of improving the learning, memory and antioxidant capacity of AD model rats to a certain extent.
