不同剂量熊果酸对糖尿病大鼠肝肾功能及血脂血糖水平的影响 被引量:6

Effects of Different Doses of Ursolic Acid on Liver and Kidney Function, Blood Lipid and Blood Glucose Level in Diabetic Rats

摘要 目的:探讨不同剂量熊果酸对糖尿病大鼠肝肾功能及血脂、随机血糖(random blood glucose,RBG)的影响。方法:将60只SD大鼠采用随机数字表法分为对照组、模型组、二甲双胍组、熊果酸低剂量组、熊果酸中剂量组和熊果酸高剂量组,每组10只。应用高脂饮食和尾静脉注射链脲佐菌素建立糖尿病大鼠模型,对照组不建立模型,二甲双胍组给予二甲双胍100 mg·kg^-1进行干预,熊果酸低剂量组、中剂量组和高剂量组分别给予熊果酸25 mg·kg^-1、50 mg·kg^-1、100 mg·kg^-1进行干预,每天灌胃给药1次,共6周。比较各组大鼠一般情况、RBG、三酰甘油(triglyceride,TG)、总胆固醇(total cholesterol,TC)、低密度脂蛋白(low density lipoprotein,LDL)、丙氨酸氨基转移酶(alanine aminotransferase,ALT)、门冬氨酸氨基转移酶(aspartate aminotransferase,AST)、总胆红素(total bilirubin,TBil)、血肌酐(serum creatinine,Scr)、血尿素氮(blood urea nitrogen,BUN)、尿酸(uric acid,UA)、肾脏系数水平。结果:其他5组大鼠精神状态、活动情况、饮食情况、排泄情况优于模型组。其他5组RBG、TG、TC、LDL、ALT、AST、Scr、BUN、UA、肾脏系数高于对照组(P<0.05);其他5组RBG、TG、TC、LDL、ALT、AST、Scr、BUN、UA、肾脏系数低于模型组(P<0.05);熊果酸低剂量组RBG、TG、TC水平高于二甲双胍组,熊果酸高剂量组TG、TC、LDL水平低于二甲双胍组,熊果酸中剂量组、高剂量组RBG水平高于二甲双胍组(P<0.05);熊果酸低剂量组、中剂量组、高剂量组RBG、TG、TC、LDL水平呈逐渐降低的趋势(P<0.05);熊果酸低剂量组、中剂量组ALT、AST、Scr、BUN、UA、肾脏系数高于二甲双胍组与熊果酸高剂量组(P<0.05);各组大鼠TBil水平比较,差异无统计学意义(P>0.05)。结论:熊果酸可以降低糖尿病大鼠RBG、血脂水平,改善肝肾功能,且100 mg·kg^-1熊果酸效果优于25 mg·kg^-1、50 mg·kg^-1。 Objective:To investigate the effects of different doses of ursolic acid on liver and kidney function,blood lipid and random blood glucose(RBG) level in diabetic rats.Methods:60 SD rats were divided into the control group,the model group,the metformin group,the low dose group,the middle dose group and the high dose group according to the random digital table method,with 10 rats in each group.The diabetic rat models were established by high fat diet and injection of streptozotocin in the tail vein.The control group did not establish model.The metformin group intervened with 100 mg·kg^-1 metformin.The low dose group,the middle dose group and the high dose group were given ursolic acid 25 mg·kg^-1,50 mg·kg^-1,100 mg·kg-1 intervention,respectively,intragastric administration once a day for 6 weeks.The general conditions of rats,RBG,triglyceride(TG),total cholesterol(TC),low density lipoprotein(LDL),alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBil),serum creatinine(Scr),blood urea nitrogen(BUN),uric acid(UA),kidney coefficient level of the rats were compared.Results:The other five groups of rats were superior to the model group in mental state,activity,diet and excretion.The other five groups of RBG,TG,TC,LDL,ALT,AST,Scr,BUN,UA and kidney coefficients were higher than those of the control group(P<0.05).The other five groups of RBG,TG,TC,LDL,ALT,AST,Scr,BUN,UA and kidney coefficients were lower than those of the model group(P<0.05).The levels of RBG,TG and TC in the low dose group were higher than those in metformin group.The levels of TG,TC and LDL in the high dose group were lower than those in metformin group,and the RBG level in the middle dose group and high dose group was higher than those in metformin group(P<0.05).RBG,TG,TC and LDL levels in low dose group,medium dose group,high dose group decreased(P<0.05);ALT,AST,Scr,BUN,UA,kidney coefficient in low dose group,middle dose group were higher than those of metformin group and high dose group(P<0.05).There was no significant difference in the TBil level of rats in each group(P>0.05).Conclusion:Ursolic acid can reduce the level of RBG and blood lipid in diabetic rats and improve the function of liver and kidney,and the effect of ursolic acid on 100 mg/kg is better than that of 25 mg/kg and 50 mg·kg^-1.
