清热解毒中药对肝癌患者血清CXCL1的影响

Chinese Medicine with Heat-clearing and Toxin-resolving Function on Serum CXCL1 in Patients with Primary Liver Cancer

目的:观察清热解毒中药对原发性肝癌患者血清CXCL1 mRNA和蛋白表达的影响。方法:60例受试者分为正常对照组(健康者24例)和肝癌组(原发性肝癌患者36例),比较两组受试者血清CXCL1 mRNA和蛋白表达情况。同时肝癌组随机分为两组:服用清热解毒中药组(19例)和未服用清热解毒中药组(17例),比较两组患者血清CXCL1 mRNA和蛋白表达情况。正常对照组与肝癌组均抽血送检,其中肝癌组中的中药组服用清热解毒中药方;通过RT-PCR、Western检测血清CXCL1的mRNA和蛋白表达,比较正常对照组与肝癌组之间的差异,进一步比较服用清热解毒中药组与未服用清热解毒中药组之间的差异,探讨清热解毒中药对CXCL1 mRNA和蛋白水平的影响。结果:①正常对照组、肝癌组两组血清CXCL1 mRNA水平比较,差异有统计学意义(P<0.01),正常对照组显著低于肝癌组;服用清热解毒中药组与未服用清热解毒中药组比较,CXCL1 mRNA水平差异有统计学意义(P<0.01),服用清热解毒中药组显著低于未服用清热解毒中药组。②正常对照组、肝癌组两组血清CXCL1蛋白水平比较,差异有统计学意义(P<0.01),正常对照组显著低于肝癌组(P<0.01);服用清热解毒中药组与未服用清热解毒中药组CXCL1蛋白水平比较,差异有统计学意义(P<0.01),服用清热解毒中药组显著低于未服用清热解毒中药组。结论:原发性肝癌患者血清CXCL1的mRNA、蛋白水平高于健康人的表达,清热解毒中药方可以显著下调肝癌患者血清中CXCL1的mRNA含量和蛋白的表达。

Objective:To observe the effect of Chinese medicine with heat-clearing and toxin-resolving function on serum CXCL1 mRNA and protein expression in patients with primary liver cancer.Methods:60 cases were divided into normal control group(24 healthy cases) and liver cancer group(36 cases of primary liver cancer patients).The serum CXCL1 mRNA and protein expression were compared between the two groups.At the same time,the liver cancer group was randomly divided into two groups:the group taking Chinese medicine with heat-clearing and toxin-resolving function(19 cases) and the group not taking Chinese medicine with heat-clearing and toxin-resolving function(17 cases).The serum CXCL1 mRNA and protein expression were compared between the two groups.Both the normal control group and the liver cancer group were given blood for examination,and the Chinese medicine group in the liver cancer group was given Chinese medicine prescription with heat-clearing and toxin-resolving function.The mRNA and protein content of serum CXCL1 were detected by RT-PCR and Western methods,and the difference between normal control group and liver cancer group was compared.To further compare the differences between the group taking Chinese medicine with heat-clearing and toxin-resolving function and the group not taking Chinese medicine with heat-clearing and toxin-resolving function,and to explore the effect of Qingrejiedu Chinese medicine on CXCL1 mRNA and protein levels.Results:①The serum CXCL1 mRNA levels in the normal control group and the liver cancer group were significantly different(P<0.01),and the normal control group was significantly lower than the liver cancer group.The difference of CXCL1 mRNA level between the group taking Chinese medicine with heat-clearing and toxin-resolving function and the group not taking Chinese medicine with heat-clearing and toxin-resolving function was statistically significant(P<0.01).The group taking Chinese medicine with heat-clearing and toxin-resolving function was significantly lower than the group not taking Chinese medicine with heat-clearing and toxin-resolving function.②There were significant differences in serum CXCL1 protein levels between the normal control group and the liver cancer group(P<0.01),and the normal control group was significantly lower than the liver cancer group(P<0.01).The difference of CXCL1 protein level between the group taking Chinese medicine with heat-clearing and toxin-resolving function and the group not taking Chinese medicine with heat-clearing and toxin-resolving function was statistically significant(P<0.01).The group taking Chinese medicine with heat-clearing and toxin-resolving function was significantly lower than the group not taking Chinese medicine with heat-clearing and toxin-resolving function.Conclusion:The mRNA and protein levels of serum CXCL1 in patients with primary liver cancer are higher than those in healthy people.Chinese medicine with heat-clearing and toxin-resolving function can significantly down-regulate the mRNA and protein expression of CXCL1 in serum of patients with liver cancer.
