分析苏木素复染对组织切片免疫组化检测结果的影响 被引量:1

摘要 目的探讨苏木素复染对组织切片免疫组化(IHC)检测结果的影响效果。方法选择本院2016年1月至2018年6月收治的肺癌患者25例作为研究对象,均抽取患者的肺癌组织进行切片后免疫组化定量分析,根据是否采取苏木素复染分为复染组与非复染组,主要测定组织切片中的CK与Ki-67表达情况,利用数码相机与采图软件于显微镜下观察组织显色反应图片,并进一步分析CK与Ki-67蛋白表达阳性单位,即PU值,采取统计学分析二者之间的差异。结果复染组与非复染组肺癌组织切片图像显示CK与Ki-67蛋白表达有着一定的差异;复染组肺癌组织切片CK蛋白表达阳性单位、Ki-67蛋白表达阳性单位与非复染组比较差异均有统计学意义(P<0.05),复染组二者均高于非复染组。结论苏木素复染对组织切片免疫组化检测结果有明显影响,会造成浆定位与核定位蛋白阳性强度显著提高,存在明显偏差,从而影响组织切片定量分析,按照这样的结果提出为了尽量组织切片免疫组化定量检测结果的准确性,不宜实施苏木素复染。 Objective To investigate the effect of hematoxylin restaining on immunohistochemical (IHC) detection of tissue sections. Methods 25 patients with lung cancer admitted in our hospital from January 2016 to June 2018 were selected for immunohistochemical quantitative analysis. According to whether hematoxylin restaining was adopted, 25 patients with lung cancer were divided into two groups: restained group and nonrestained group. The expression of CK and Ki-67 in tissue sections was determined. The tissue color reaction images were observed under microscope with digital camera and image acquisition software, and the positive units of CK and Ki-67 protein, PU, were further analyzed. Value, statistical analysis of the difference between the two. Results The expression of CK and Ki-67 protein was significantly different between the restained group and the non-restained group. The expression of Ki-67 protein of CK protein was significantly higher in the restained group than that in the nonrestained group. The expression of Ki-67 protein in the restained group was higher than that in the non-restained group (P<0.05). Conclusion Hematoxylin restaining has a significant effect on the immunohistochemical results of tissue sections, which will result in a marked increase in the positive intensity of cytoplasmic and nuclear localization proteins and a significant deviation, thus affecting the quantitative analysis of tissue sections. The results suggested that hematoxylin restaining should not be carried out in order to maximize the accuracy of immunohistochemical quantitative detection of tissue sections.
