摘要
目的:探索建立一种右心室重构动物模型的方法。方法:取健康成年绵羊13只,分为实验组和对照组,实验组羊接受外科手术,进行肺动脉束扎术,束扎后使肺动脉跨瓣压差>60 mmHg(1 mmHg=0.133 kPa),增加右心室后负荷,引起代偿性右心室结构及功能的改变,诱发羊右心室重构。对照组仅接受开胸手术。术前、术后3个月、6个月分别对两组行超声心动图检查监测心室功能、心室壁厚度、心室腔大小及心室容积的变化,并进行统计学分析。术后6个月剪取两组右心室前壁组织制成HE染色切片标本进行病理学观察分析。结果:本文通过羊肺动脉束扎术建立右心室重构的动物模型,通过6个月监测发现,右心功能明显减退,右心室前壁厚度增加,右心室腔直径、容积明显大于对照组,右心室壁心肌病理切片显示出现明显的右心室重构病理变化。结论:此方法可较好地建立右心室重构动物模型。
Objective: Explore ways to establish an animal model of right ventricular remodeling. Methods: Thirteen healthy adult cotton sheep were divided into experimental group and control group. The experimental group received surgery for pulmonary artery ligation. After bundled, the pulmonary artery transvalvular pressure difference was greater than 60 mmHg, and the right ventricular afterload was increased, causing compensation. Changes in the structure and function of the right ventricle induce cervical right ventricular remodeling. The control group only received thoracotomy. The changes of ventricular function, ventricular wall thickness, ventricular cavity size and ventricular volume were measured by echocardiography in the experimental group and the control group before and 3 months and 6 months after operation, and statistical analysis was performed. At 6 months after operation, the right ventricular anterior wall tissue of the experimental group and the control group were cut into HE stained sections for pathological analysis. Results: In this paper, an animal model of right ventricular remodeling was established by sheep pulmonary artery ligation. After 6 months of monitoring, the right ventricular function was significantly reduced, the right ventricular anterior wall thickness was increased, and the right ventricular cavity diameter and volume were significantly larger than the control group, right ventricle. Myocardial pathological sections showed significant pathological changes in right ventricular remodeling. Conclusions: This method establishes an animal model of right ventricular remodeling.
