摘要
目的:研究单羰基姜黄素类似物EF24对胃癌SGC-7901细胞凋亡和焦亡的影响。方法:常规培养胃癌BGC-823和SGC-7901细胞,给予不同浓度的EF24处理细胞,MTT法检测胃癌细胞的活性,细胞集落法检测EF24对SGC-7901细胞集落形成的影响,流式细胞术分别检测EF24对SGC-7901细胞凋亡的影响和细胞内活性氧簇(ROS)含量变化对SGC-7901细胞生长抑制情况的影响,Western Blotting法检测EF24作用SGC-7901细胞对Bcl-2和Bax蛋白的影响,显微成像初步验证EF24通过升高ROS对SGC-7901细胞焦亡的影响。结果:与对照组比较,EF24呈浓度依赖抑制胃癌细胞生长(P<0.05或P<0.01),对BGC-823和SGC-7901胃癌细胞的半抑制浓度(IC50)分别为4.35μmol·L^-1和4.96μmol·L^-1;EF24呈浓度依赖性抑制SGC-7901胃癌细胞集落形成,促进细胞凋亡和促凋亡蛋白Bax的表达,抑制抑凋亡蛋白Bcl-2的表达(P<0.05或P<0.01);EF24升高SGC-7901胃癌细胞内ROS的含量并诱导细胞凋亡和焦亡(P<0.05或P<0.01)。结论:EF24能够抑制胃癌细胞生长,升高SGC-7901细胞的ROS含量,从而诱导细胞凋亡和焦亡。
Objective:To investigate the inhibitory effect of monocarbonyl curcumin analogue EF24 on the apoptosis and pyroptosis of SGC-7901 gastric cancer cells.Methods:BGC-823 and SGC-7901 gastric cancer cells were cultured by the conventional method,and EF24 treatment groups at different concentrations were set up.MTT assay was used to evaluate the activity of gastric cancer cells,and cell colony assay was performed for the effects on the colony formation of SGC-7901 cells.The SGC-7901 cells apoptosis and the growth inhibition of SGC-7901 cells via the promotion of ROS levels were evaluated by flow cytometry.Western blot was used to detect the expressions of Bax and Bcl-2 proteins after treated with EF24.The effect of EF24 on the ROS-based SGC-7901 cells pyroptosis was confirmed by microscopic imaging.Results:Compared with the control group,EF24 inhibited gastric cancer cells in a concentrationdependent manner(P<0.05 or P<0.01)with the IC50 value on BGC-823 and SGC-7901 gastric cancer of 4.35μmol·L^-1and 4.96μmol·L^-1,respectively.EF24 restrained the SGC-7901 colony formation in a concentration-dependent pattern and promoted SGC-7901 cells apoptosis.EF24 promoted the expression of the pro-apoptotic protein Bax and inhibited the expression of the anti-apoptotic protein Bcl-2 in a concentration-dependent manner(P<0.05 or P<0.01).EF24 induced SGC-7901 cells apoptosis and pyroptosis by increasing intracellular ROS content(P<0.05 or P<0.01).Conclusion:EF24 can inhibit the growth of gastric cancer cells,and induce the apoptosis and pyroptosis of SGC-7901 cancer cells by promoting the level of intracellular ROS.
