摘要
目的:优化绞股蓝总皂苷提取工艺,获取绞股蓝总皂苷最佳提取工艺条件及技术参数。方法:采用Box-Benhnken中心组合设计,以提取物中总皂苷含量和总皂苷提取率的总权重评分值为评价指标,对乙醇浓度、浸渍时间、渗漉流速3个影响因素进行考察,以Design-Expert 8.05软件对试验数据作统计处理,优选确定最佳提取条件。结果:优化得到最佳工艺条件为:乙醇浓度为72%,浸渍时间为60 h,渗漉流速为3.0ml·min^-1。结论:优选得到的工艺稳定、可行,可作为绞股蓝总皂苷提取提供参考依据。
Objective:To optimize the extraction technique of total ginsenoside in gynostemma pentaphyllum.makino to obtain the optimal extraction conditions and parameters.Methods:Central composite design-response surface methodology was used with the total weight score of the total ginsenoside content and yield as the evaluation index.Three factors including ethanol concentration,immersion time and percolation velocity were designed,and Design-Expert 8.05 software was used for the statistical processing of the experimental data.Results:The optimal extraction conditions of gynostemma pentaphyllum.makino were as follows:the ethanol concentration was72%,the immersion time was 60 hours and the percolation velocity was 3.0 ml·min^-1.Conclusion:The application of central composite design-response surface methodology in the extraction optimization of total ginsenoside in gynostemma pentaphyllum.makino is stable and feasible,and the conditions can provide theoretical guidance for the extraction.
