HPLC同时测定鹿衔草提取物中香草酸、金丝桃苷、槲皮素-3-O-呋喃阿拉伯糖苷、槲皮素和梅笠草素的含量

Simultaneous Determination of Vanillic Acid,Hyperin,Quercetin-3-O-furanoarabinoside,Quercetin and Chimaphilin in the Extracts of Pyrolae Herba by HPLC

目的:建立HPLC同时测定鹿衔草提取物中香草酸、金丝桃苷、槲皮素-3-O-呋喃阿拉伯糖苷、槲皮素和梅笠草素的含量。方法:色谱柱:Zorbax SB-C18(250 mm×4.6 mm,5μm)柱,流动相:乙腈-0.1%醋酸水,梯度洗脱;柱温:35℃,流速:1.0 ml·min^-1,检测波长:255 nm。结果:香草酸、金丝桃苷、槲皮素-3-O-呋喃阿拉伯糖苷、槲皮素和梅笠草素分别在3.71~148.40μg·ml^-1(r=0.999 9)、5.92~236.70μg·ml^-1(r=0.999 8)、3.36~134.25μg·ml^-1(r=0.999 8)、3.94~157.80μg·ml^-1(r=0.999 9)和3.10~124.15μg·ml^-1(r=0.999 8)范围内线性关系良好,平均回收率分别为99.01%、98.80%、98.48%、98.71%和98.62%,RSD分别为0.95%、1.15%、1.36%、0.94%和1.02%(n=6)。结论:该方法快捷简便、准确性及重复性均良好,可用于鹿衔草提取物及相关制剂质量的评价和控制。

Objective:To set up an HPLC method for the simultaneous determination of vanillic acid,hyperin,quercetin-3-O-furanoarabinoside,quercetin and chimaphilin in the extracts of Pyrolae Herba.Methods:A Zorbax SB-C18column(250 mm×4.6 mm,5μm)was used and use mobile phase was acetonitrile-0.1%acetic acid with gradient elution at a flow rate of 1.0 ml·min^-1.The column temperature was 35℃.The detection wavelength was 255 nm.Results:Good linear relationships were found within the range of 3.71-148.40μg·ml-1(r=0.999 9)for vanillic acid,5.92-236.70μg·ml^-1(r=0.999 8)for hyperin,3.36-134.25μg·ml^-1(r=0.999 8)for quercetin-3-O-furanoarabinoside,3.94-157.80μg·ml^-1(r=0.999 9)for quercetin and 3.10-124.15μg·ml^-1(r=0.999 8)for chimaphilin.The average recovery was 99.01%(RSD=0.95%),98.80%(RSD=1.15%),98.48%(RSD=1.36%),98.71%(RSD=0.94%)and 98.62%(RSD=1.02%)(n=6),respectively.Conclusion:The method is rapid,simple,accurate and reproducible,and can be used for the quality evaluation and control of Pyrolae Herba extracts and the related preparations.
