摘要
以黄果、满天星和紫果3个不同品种百香果果皮和果肉为材料,采用2代Illumina高通量测序技术进行转录组测序,获得黄果果皮47 875 830条、果肉44 325 678条,满天星果皮47 019 772条、果肉45 472 434条,紫果果皮48 789 846条、果肉49 372 968条Clean reads片段,包含黄果果皮7 181 374 500 nt、果肉6 648 851 700 nt,满天星果皮7 052 965 800 nt、果肉6 820 865 100 nt,紫果果皮7 318 476 900 nt、果肉7 405 945 200 nt核苷酸序列信息;经拼接组装,分别获得平均长度为1 487 nt的黄果34 968条Unigene片段,平均长度1 361 nt的满天星41 896条Unigene片段和平均长度1 329 nt的紫果41 701条Unigene片段。GO富集分析表明,在生物过程、细胞组分和分子功能3大类别上,黄果、满天星和紫果间均分布有差异基因。KEGG富集分析表明,黄果与满天星在细胞胞吞作用过程中存在较大差异,黄果与紫果在玉米素生物合成和糖酵解途径中差异基因富集显著,满天星与紫果在缬氨酸、亮氨酸和异亮氨酸生物合成中存在较大差异。
The Illumina high-throughput sequencing technology was used to establish three transcriptome libraries by used the RNA pools which were isolated from peel and flesh of Passiflora edulia Sims cv.‘Gold’, Passiflora edulia Sims cv.‘Mantianxing’ and Passiflora edulia Sims cv.‘Purple’ respectively. Anaverage length of 150 nt and the total of 47 875 830 and 44 325 678 clean reads for peel and flesh of Passiflora edulia Sims cv.‘Gold’, 47 019 772 and 45 472 434 clean reads for peel and flesh of Passiflora edulia Sims cv.‘Mantianxing’, 48 789 846 and 49 372 968 clean reads for peel and flesh of Passiflora edulia Sims cv.‘Purple’ were separately generated, it contains the following nucleotide sequence information:7 181 374 500 nt in Passiflora edulia Sims cv.‘Gold’ peel, 6 648 851 700 nt in Passiflora edulia Sims cv.‘Gold’ flesh, 7 052 965 800 nt in Passiflora edulia Sims cv.‘Mantianxing’ peel, 6 820 865 100 nt in Passiflora edulia Sims cv.‘Mantianxing’ flesh, 7 318 476 900 nt in Passiflora edulia Sims cv.‘Purple’ peel, 7 405 945 200 nt in Passiflora edulia Sims cv.‘Purple’ flesh. According to the assembled by splicing, it also respectively produced 34 968 Unigene with a mean length 1 487 nt for Passiflora edulia Sims cv.‘Gold’, 41 896 Unigene with a mean length 1 361 nt for Passiflora edulia Sims cv.‘Mantianxing’ and 41 701 Unigene with a mean length 1 329 nt for Passiflora edulia Sims cv.‘Purple’. GO enrichment analysis showed that there were differentially distributed genes in the three categories(Passiflora edulia Sims cv.‘Gold’, Pas siflora edulia Sims cv.‘Mantianxing’and Passiflora edulia Sims cv.‘Purple’)of biological processes,cellular components and molecular function.The analysis with the KEGG enrichment showed that there were significant differences between Passiflora edulia Sims cv.‘Gold’and Passiflora edulia Sims cv.‘Mantianxing’in the process of the Phagosome pathway,and the Zeatin biosynthesis pathway and Glycolysis/Gluconeogenesis pathway in Passiflora edulia Sims cv.‘Gold’had significant difference compared to Passiflora edulia Sims cv.‘Purple’,and the Valine,leucine and isoleucine biosynthesis pathway in Passiflora edulia Sims cv.‘Mantianxing’had significant difference compared to Passiflora edulia Sims cv.‘Purple’.
