黄芪注射液联合葛根素注射液对2型糖尿病小鼠心肌的保护作用及其机制 被引量:17

摘要 目的:探讨黄芪注射液联合葛根素注射液对2型糖尿病小鼠心脏的保护作用及其机制。方法:将造模成功的2型糖尿病KKAy小鼠随机分为模型组和治疗组(黄芪注射液联合葛根素注射液腹腔注射),同周龄雄性KKAy小鼠作为正常对照组,并于21、24和28周龄时分别处死小鼠。HE染色观察心肌组织形态变化;TUNEL染色法检测心肌细胞凋亡;real-time PCR检测小鼠心脏组织中葡萄糖调节蛋白78(GRP78)、C/EBP同源蛋白(CHOP)和p53上调凋亡调控因子(PUMA) mRNA的表达;Western blot检测小鼠心脏组织中GRP78、CHOP、PUMA、caspase-3、cleaved caspase-3、caspase-9、cleaved caspase-9、多腺苷二磷酸核糖聚合酶(PARP)和cleaved PARP蛋白的表达。结果:模型组小鼠心肌细胞发生肥大,可见到部分细胞出现肌浆溶解和坏死,治疗组小鼠病变减轻;与对照组比较,模型组小鼠心肌细胞凋亡数显著增加(P<0.01),治疗组小鼠心肌细胞凋亡数较模型组显著降低(P<0.05);模型组小鼠21、24和28周龄心肌组织中GRP78、CHOP和PUMA mRNA和蛋白水平,以及cleaved caspase-3、cleaved caspase-9和cleaved PARP蛋白表达水平与正常对照组比较显著增加(P<0.01);治疗组小鼠心肌组织中GRP78、CHOP和PUMA mRNA和蛋白水平以及cleaved caspase-3、cleaved caspase-9和cleaved PARP蛋白表达水平均显著低于模型组(P<0.01)。结论:黄芪注射液联合葛根素注射液对2型糖尿病小鼠心肌具有保护作用,其机制可能与抑制内质网应激和caspase通路的活化,从而抑制细胞凋亡有关。 AIM: To investigate the effect of Huangqi injection combined with puerarin injection on the myocardium of the mice with type 2 diabetes. METHODS: Diabetic KKAy mice were randomly divided into model group and treatment group(Huangqi injection combined with puerarin injection). The male KKAy mice of the same age were used as control group. All mice were sacrificed at 21, 24 and 28 weeks. Morphological changes of the myocardium were observed by HE staining. Apoptosis of the cardiomyocytes was measured by TUNEL staining. The mRNA levels of glucose-regulated protein 78(GRP78), C/EBP hoinologous protein(CHOP) and p53 up-regulated modulator of apoptosis(PUMA) were detected by real-time PCR, and the protein levels of GRP78, CHOP, PUMA, caspase-3, cleaved caspase-3, caspase-9, cleaved caspase-9, poly(ADP-ribose) polymerase(PARP) and cleaved PARP were determined by Western blot. RESULTS: Cardiomyocyte hypertrophy, partly dissolved sarcoplasm and necrosis were observed in model group, and these lesion were alleviated in treatment group. Obvious increased apoptosis in model group and significantly decreased apoptosis of cardiomyocytes in treatment group was observed(P<0.05). At 21, 24 and 28 weeks, the mRNA and protein levels of GRP78, CHOP and PUMA and the protein levels of cleaved caspase-3, cleaved caspase-9 and cleaved PARP in model group were increased significantly as compared with control group(P<0.01), and these in treated group were decreased compared with model group. CONCLUSION: Huangqi injection combined with puerarin injection has cardioprotective effects on type 2 diabetes mice and its mechanism of the action was implemented via inhibiting the activation of endoplasmic reticulum stress and caspase pathway, thus resulting in suppressed apoptosis.
