摘要
目的对乌甘胶囊主要成分进行定性定量方法研究。方法采用显微鉴别法定性鉴别处方中的海螵蛸和甘草;采用TLC法定性鉴别处方中甘草;采用HPLC法梯度洗脱测定甘草中甘草苷的含量,色谱柱为WondaSil C18(4.6 mm×250 mm,5μm),以乙腈为流动相A、0.1%磷酸溶液为流动相B进行梯度洗脱,检测波长276 nm,流速1.0 ml/min,柱温25℃。结果显微鉴别法结果显示,处方中的海螵蛸、甘草显微特征明显;TLC法结果显示,甘草斑点清晰,分离效果好;HPLC法结果显示,甘草苷的进样量在0.041 23~0.453 51μg范围内与峰面积呈良好的线性关系(r=1),平均加样回收率为98.61%,RSD为1.51%(n=6)。结论本实验方法可作为乌甘胶囊的定性定量控制方法。
Objective To conduct the qualitative and quantitative analysis of Wu Gan Capsule.Methods Cuttlebone and glycyrrhiza uralensis in Wu Gan Capsule were identified by microscopic identification.Glycyrrhiza ura-lensis was identified by TLC.The content of liquiritin of glycyrrhiza uralensis in Wu Gan Capsule was determined by gradient elution of HPLC.WondaSil C18(4.6 mm×250 mm, 5 μm) column was used;the mobile phase of gradient elution consisted of acetonitrile(A) and 0.1% phosphoric acid(B);UV detection wavelength was set at 276 nm;the column temperature was 25 ℃;the flow rate was 1.0 ml/min.Results There were obvious microscopic characte-ristics of cuttlebone and glycyrrhiza uralensis in Wu Gan Capsule according to the results of microscopic identification method.TLC method showed good resolution effect with clear spots of glycyrrhiza uralensis.The good linearity was obtained within the range of 0.041 23~0.453 51μg(liquiritin,r=1) by HPLC,and the average recovery rate was 98.61%.Conclusion The established method can be applied in the quality control of Wu Gan Capsule.
