含铬革屑碱-酸水解制备氨基酸液工艺探究 被引量:4

摘要 本课题主要以含铬革屑为原料用NaOH、CaO混合碱进行脱铬提取大分子胶原蛋白,再用酸水解制备氨基酸液,并以水解度作为指标,系统考察了HCl、H2SO4和HNO3酸解过程中水解温度、水解时间、酸用量对水解效果的影响,确定最佳水解条件。实验结果表明:HCl水解体系最佳水解条件为90℃、6h、盐酸用量为2.5mol/L,水解度为47.85%;H2SO4水解体系最佳水解条件为90℃、10h、硫酸用量为2.5mol/L,水解度为49.39%;HNO3水解体系最佳水解条件为100℃、12h、硝酸用量为2.0mol/L,水解度为40.68%。 In this paper, the macromolecule collagen was extracted by dechroming with NaOH and CaO mixed as base, and the amino acid solution was prepared by acid hydrolysis. The degree of hydrolysis was used as the index, and the optimum hydrolysis conditions were determined by the effect of hydrolysis temperature, hydrolysis time and acid dosage on the hydrolysis effect of HCl, H2SO4 and HNO3. Experimental results show that: the optimum hydrolysis conditions for HCl hydrolysis are 90℃,6 h, the dosage of HCl is 2.5 mol/L, the hydrolysis degree is 47.85%;The optimum hydrolysis conditions for H2SO4 hydrolysis are 90℃, 10 h, the dosage of H2SO4 is 2.5 mol/L, and the degree of hydrolysis are 49.39%;the optimum hydrolysis conditions for HNO3 hydrolysis are 100℃, 12 h, the dosage of HNO3 is 2.0 mol/L and the degree of hydrolysis is 40.68%.
