安罗替尼对肺癌A549细胞增殖、迁移侵袭的影响

Effect of Anlotinib on the Proliferation and Migration of Lung Cancer A549 Cells

目的研究安罗替尼对肺癌A549细胞增殖和迁移侵袭的影响。方法采用不同浓度的安罗替尼作用于肺癌A549细胞,利用CCK8法检测细胞增殖能力;流式细胞术检测A549细胞凋亡情况;通过划痕实验测定各组细胞的迁移能力;Transwell实验检测细胞侵袭能力。结果安罗替尼处理后,CCK8结果显示A549细胞的增殖能力降低(P<0.05;划痕实验表明A549细胞迁移率降低(P<0.05;Transwell实验观察到穿过Transwell小室膜的A549细胞数减少(P<0.05),侵袭能力受抑制;凋亡双染法发现,A549细胞凋亡比例增高(P<0.05)。结论安罗替尼能够抑制肺癌A549细胞增殖、迁移侵袭,促进肺癌A549细胞凋亡。

Objective To study the effect of anlotinib on the proliferation and migration of lung cancer A549 cells.Methods The lung cancer A549 cells were treated with different concentrations of anlotinib.The CCK8 method was used to detect the cell proliferation, the flow cytometry was performed to detect the apoptosis of A549 cells, the scratch test was adopted to determine their migration ability, and the Transwell test was employed to detect their invasion ability.Results After the treatment with anlotinib, the CCK8 results showed that the proliferation of A549 cells was significantly reduced ( P <0.05).The scratch test showed that the migration rate of A549 cells was significantly decreased ( P <0.05).The Transwell test showed a significant decrease in the number of A549 cells passing through the Transwell chamber membrane ( P <0.05) and an inhibited invasion ability.The apoptotic double staining showed that the apoptotic rate of A549 cells increased significantly ( P <0.05).Conclusion Anlotinib inhibits the proliferation, migration and invasion of lung cancer A549 cells and promotes their apoptosis.
