摘要 目的优化验证天麻素的提取工艺。方法用醇提法从天麻中提取分离天麻素,高效液相色谱法测定天麻素含量。以天麻素提取率为评价指标,用单因素法考察各试验因素对天麻素提取率的影响;用正交试验法优化提取工艺参数。结果单因素法考察提取工艺条件为:料液比1∶18,醇提浓度70%,提取次数4次,提取时间1h。正交试验优化提取工艺条件为:料液比1∶16,醇提浓度65%,醇提次数4次,提取时间1.5h,天麻素提取率为0.983%。结论经优化的天麻素提取工艺稳定可靠。 Objective To optimize and verify the extraction technology of gastrodin.Methods Gastrodin was extracted and separated from Gastrodia Rhizoma by the alcohol extraction method and the gastrodin content was determined by high performance liquid chromatography.With the extraction rate of gastrodin as the evaluation index, the influence of each test factor on the extraction rate of gastrodin was investigated by the single factor method and the extraction technology parameters were optimized by the orthogonal test.Results The conditions of the extraction technology in the single factor method were as follows: solid-liquid ratio of 1 ∶18, alcohol extraction concentration of 70%, 4 times of extraction and 1 hour of extraction.Those of the optimized extraction technology in the orthogonal test were as follows: solid-liquid ratio of 1 ∶16, alcohol extraction concentration of 65%, 4 times of extraction and 1.5 hour of extraction, and the extraction rate of gastrodin was 0.983%.Conclusion The optimized extraction technology of gastrodin is stable and reliable.
