摘要
目的观察淫羊藿女贞子配伍协同激素对哮喘大鼠肺组织细胞凋亡的影响并探讨其作用机制。方法SD雄性大鼠,随机分为7组,分别是正常对照组(对照组)、哮喘模型组(模型组)、布地奈德组、淫羊藿女贞子煎剂组(煎剂组)、布地奈德合煎剂组(合煎剂组)、淫羊藿女贞子提取物组(提取物组)、布地奈德合提取物组(合提取物组)。卵蛋白致敏及激发建立大鼠哮喘模型,药物治疗4周。TUNEL法检测肺组织细胞凋亡水平,荧光实时定量PCR检测肺组织Caspase-9、Fas、Hras、NF-κB、Xiap mRNA表达。结果与对照组比较,模型组大鼠肺组织凋亡水平及Caspase-9 mRNA表达显著下调(P <0.01),NF-κB和Xiap m RNA显著升高(P <0.01),Fas及Hras mRNA差异无统计学意义(P> 0.05)。与模型组比较,各给药组均能显著上调细胞凋亡水平(P <0.05或P <0.01),抑制NF-κB mRNA的表达(P <0.05或P <0.01);除布地奈德组外其余4个给药组可上调Xiap mRMA的表达(均P <0.05或P <0.01);煎剂组、合煎剂组、提取物组Caspase-9、Fas mRNA表达均显著差异(P <0.05或P <0.01)。结论上调哮喘大鼠肺组织凋亡水平,调节Caspase-9、Fas、NF-κB、Xiap mRNA的表达可能是淫羊藿女贞子协同激素治疗哮喘的作用机制之一。
Objective:To study the effects of Herba Epimedii,Fructus Ligustri Lucid and Budesonide on cell apoptosis in lung tissue of asthmatic rats. Methods:Male SD rats were randomly divided into 7 groups. The rat asthma model was established by ovalbumin sensitization(OVA),and the drug treatment lasted for 4 weeks. TUNEL assay was used to detect the apoptotic levels of lung cells,real-time fluorescent quantitative PCR to detect the mRNA expression of Caspase-9,Fas,Hras,NF-κB and Xiap in lung tissues. Results:Compared with Nc group,the level of apoptosis and Caspase-9 mRNA were significantly decreased(P < 0.01);mRNA of NF-κB and Xiap were significantly higher in Mo group(all P < 0.01);Fas and Hras mRNA was not statistically significant. Compared with Mo group,the level of apoptosis was significantly increased(P < 0.05 or P < 0.01) and NF-κB mRNA was significantly decreased(P < 0.05 or P < 0.01) in each drug administration group;Xiap mRNA were significantly higher in the four groups except Bun group(P < 0.05 or P < 0.01). There were statistically differences in Caspase-9 and Fas mRMA among HF1 group,Bun&HF1 group and Bun&HF2 group. Conclusion:Up-regulation of apoptotic level of lung tissue and expression of Caspase-9,Fas,NF-κB and Xiap in asthmatic rats may be one of the mechanisms of synergistic hormone therapy of Herba Epimedii and Ligustrum Lucidum.
