稳斑汤对ApoE基因敲除小鼠AS不稳定斑块凋亡相关基因Caspase-3表达的影响

Effect of Wenban Decoction on Apoptosis-related gene Caspase-3 Expression of Unstable Plaque in ApoE Knockout Mice with AS

目的观察稳斑汤对ApoE基因敲除小鼠动脉粥样硬化不稳定斑块凋亡相关基因半胱氨酸蛋白酶-3(Caspase-3)表达的影响。方法利用6~8周龄ApoE基因敲除小鼠建立动脉粥样硬化不稳定斑块模型,并随机分为空白对照组,模型组,稳斑汤低、中、高剂量组,他汀组。空白对照组及模型组均给予0.9%氯化钠溶液灌胃,稳斑汤低、中、高剂量组及他汀组分别给予不同剂量稳斑汤及阿托伐他汀钙片干预。采用HE染色进行主动脉组织病理学观察,利用反转录PCR法检测Caspase-3mRNA水平。结果与空白对照组比较,模型组Caspase-3mRNA表达水平均升高(P<0.05);与模型组比较,稳斑汤中、高剂量组Caspase-3mRNA表达水平均降低(P<0.05);与他汀组比较,稳斑汤中、高剂量组Caspase-3mRNA表达水平无明显差异(P>0.05)。镜下观察斑块面积,模型组小于稳斑汤各剂量组和他汀组。结论稳斑汤可能通过下调ApoE基因敲除小鼠AS不稳定斑块凋亡相关基因Caspase-3的表达来干预动脉粥样硬化不稳定斑块的形成及破裂。

Objective:To observe the effect of compound Wenban Decoction on Caspase-3 mRNA expression of unstable plaque in ApoE knockout mice with Atherosclerosis. Methods:A number of 6 to 8 weeks old ApoE knockout mice were selected to establish AS unstable plaque model and randomly divided into the blank control group,the model group,the statin group and Wenban Decoction low,medium and high dose groups. They were given normal saline,low dose,medium dose,high dose compound Wenban Decoction and Atorvastatin. Aortic tissue were observed under optical microscope pathology after HE staining. The mRNA expression levels of Caspase-3 were detected with RT-PCR. Results:Compared with the blank control group,Caspase-3 mRNA expression levels were higher(P < 0.05);compared with model group,Caspase-3 mRNA expression levels in middle and high dose Wenban Decoction groups were lower(P < 0.05). Compared with the statin group,there were no difference in middle and high dose Wenban Decoction groups on expression levels of Caspase-3 mRNA(P > 0.05). The plaque area in different dose groups and the statin group were smaller than that of the model group under light microscope. Conclusion:Wenban Decoction may intervene the formation and eruption of AS unstable plaque through down-regulation the expressions of Caspase-3 mRNA in ApoE knockout mice AS unstable plaque.
