摘要
目的观察枳椇饮对急性酒精中毒小鼠行为活动的影响,验证其醒酒的疗效。方法将50只昆明小鼠(雌雄各半)随机分为模型组、枳椇饮低剂量组、中剂量组、高剂量组和阳性对照组,每组10只;各组均以10mL/kg为标准给小鼠灌酒,灌酒后3个浓度枳椇饮组以不同浓度的枳椇饮灌胃,模型组以相同剂量给0.9%氯化钠注射液,对照组给牡蛎大豆肽肉碱口服液。给药剂量分别为:枳椇饮低剂量组、中剂量组、高剂量组分别灌胃给药0.25g/kg、0.5g/kg和1g/kg枳椇饮。牡蛎大豆肽肉碱口服液对照组给予10mL/kg牡蛎大豆肽肉碱口服液。记录小鼠从灌酒到翻正反射消失的潜伏时间和翻正反射的恢复时间以及5、30和150min自主活动次数,然后进行统计学分析。结果与模型组比较,枳椇饮高剂量药物干预组显著提高了翻正反射消失潜伏时间(P<0.05);与模型组比较,阳性组、枳椇饮低、高剂量组翻正反射的恢复时间显著缩短。5min时,与模型组比较,枳椇饮高剂量组自主活动显著减少(P<0.05);30min时,与模型组比较,各组实验小鼠自主活动差异无统计学意义(P>0.05)。150min时,与模型组比较,阳性组、枳椇饮低、中、高剂量组自主活动显著增加(P<0.01)。结论枳椇饮能显著缩短急性酒精中毒小鼠翻正反射的时间,表明枳椇饮能对抗酒精所致中枢神经系统抑制进行;枳椇饮能减少兴奋期小鼠自主活动的次数,说明枳椇饮能对抗酒精中毒兴奋期的兴奋作用,在昏迷期增加小鼠的自主活动次数,表明枳椇饮能改善急性酒精中毒小鼠昏睡期的中枢抑制状态。
objective:To observe the effect of Zhiju Decoction on the activity of acute alcohol poisoning mice,and to verify the effect of sobriety on Zhiju Decoction. Methods:50 kunming mice(male and female) were randomly divided into the model group,the Zhiju Decoction low dose group,the medium dose group,the high dose group and the positive control group,10 mice in each group. The mice in each group were treated with 10 mL/kg as the standard. The hovenia acacia drink group was treated with different concentrations of hovenia acacia drink. The model group was treated with the same dose of normal saline,and the control group was treated with oyster soybean carnitine oral solution. The dosages were:Zhiju Decoction low dose group,Zhiju Decoction medium dose group,Zhiju Decoction high dose group were given 0.25 g/kg,0.5 g/kg and 1 g/kg respectively by gavage. The oral liquid of oyster soybean peptide carnitine was administered to the control group with 10 mL/kg oral liquid of oyster soybean peptide carnitine. The incubation time of mice from drinking to disappearance of righting reflex and recovery time of righting reflex,as well as The Times of autonomic activities at 5,30 and 150 min were recorded,and then statistical analysis was conducted. Results:compared with the model group,the Zhiju Decoction high-dose intervention group significantly increased the Zhiju Decoction latency (P < 0.05). Compared with the model group,the restoration time of the Zhiju Decoction and the Zhiju Decoction low and high dose groups was significantly shortened. At 5 min,compared with the model group,the Zhiju Decoction high dose group significantly decreased the independent activity (P < 0.05). At 30 min,there was no significant difference in the autonomic activity between the two groups compared with the model group. At 150 min,compared with the model group,the activity of the active group,the hovenia raisin sin(Zhiju Decoction) low,medium and high dose groups increased significantly(P < 0.01). Conclusion:Zhiju Decoction can significantly shorten the time of righting reflex of acute alcohol poisoning mice,suggesting Zhiju Decoction can resist the inhibition of alcohol induced central nervous system. Zhiju Decoction decreased the number of independent activities of the mice in the active phase,suggesting that Zhiju Decoction can resist the excitatory effect of the active phase of alcohol intoxication,and increase the number of independent activities of the mice in the coma phase,suggesting that Zhiju Decoction can improve the central inhibitory state of the mice in the sleeping phase of acute alcohol intoxication.
