摘要
为精制枸杞多糖(Lyeium barbarum Polysaccharide,LBP),并获得可靠的工艺参数,为工业化生产的实现提供必要的数据积累和依据。本文采用高速离心和膜技术相结合的试验方法对枸杞水提液进行处理,选用截留分子量为 1×10^4的超滤膜浓缩纯化 LBP,并用硫酸-苯酚显色法测定截留液和透过液多糖的含量。结果表明:操作时间和操作压力对膜通量有较大的影响。在料液温度 25 ℃、操作压力 0.8 MPa 条件下,对枸杞水提的第一汁、第二汁和合并液超滤 4~5 h,所得截留液干燥快速,其多糖含量分别为 15.51%、8.36%和 14.24%。因此,采用膜技术分离枸杞水提液的工艺操作简单,能耗小,纯化效果较显著,所得成品性状好,适用于工业化生产。
TO extract Polysaccharides from Lycium barbarum and get reliable operation parameters which could provide an essential basis and data accumulation for the industrial-scale production, Combining with the high speed centrifugation method, ultrafiltration membrane was carried out to separate and purify Lycium barbarum polysaccharides (LBP). The molecular weight cut-off of LBP was 1×10^4. Sulphuric acid-phenol method was used to determine the content of polysaccharides from the trapped fluid and permeation fluid in experimental process. Results showed that the pressure, time and temperature had great influences on the flux. While the operating pressure was 0.8 MPa, the solution temperature was 25 ℃ and the time was 4~5 hours, the content of LBP from the different rapped fluid of first juice extract, second juice extract and mixed extract were 15.51%, 8.36% and 14.24% respectively. And the trapped fluid could be quickly dried. The membrane separation technology for L. barbarum water extract with low energy consuming was simple and purification efficiency was fairly significant. The finished products had good character, suitable for industrial production.
