黄芪黄酮对氧糖剥夺/复氧复糖PC12细胞的保护作用 被引量:2

摘要 目的:研究黄芪黄酮对氧糖剥夺/复氧复糖PC12细胞的作用。方法:取对数期PC12细胞,随机分为5组:正常组、模型组(氧糖剥夺/复氧复糖组)、黄芪黄酮低(0.001mg/mL)、中(0.01mg/mL)、高(0.1mg/mL)剂量组。其中,模型组和黄芪黄酮低、中、高剂量组氧糖剥夺2h后复氧复糖24h,黄芪黄酮低、中、高剂量组在复氧复糖的同时予以黄芪黄酮不同剂量处理。用倒置相差显微镜观察PC12细胞的形态变化,MTT法和CCK-8法检测细胞的存活率。结果:正常组细胞状态良好,贴壁生长,细胞突起明显,各个突起之间交织在一起,细胞整体折光性较强,细胞表面光滑;与正常组相比,模型组部分细胞漂浮,突起收缩,细胞折光性差,细胞存活率明显降低(P<0.05);与模型组相比,黄芪黄酮低、中、高剂量组细胞状态均有明显好转,突起可见,细胞存活率均有升高(P<0.05);与黄芪黄酮低剂量组相比,黄芪黄酮中、高剂量组细胞状态较好,细胞存活率升高(P<0.05);黄芪黄酮中剂量组与高剂量组之间差异不明显(P>0.05)。结论:黄芪黄酮可改善氧糖剥夺/复氧复糖PC12细胞的生长状态,减轻细胞损伤,提高细胞存活率,发挥保护作用,且黄芪黄酮中、高剂量组的保护作用优于低剂量组。 Objective: To study the effects of Astragalus flavonoids on oxygen-glucose deprivation/reoxygenation (OGD/R) PC12 cells. Methods: Logarithmic PC12 cells were randomly divided into five groups: normal group, model group (OGD/R group), Astragalus flavonoids low (0.001 mg/mL), medium (0.01 mg/mL), high (0.1 mg/mL) dose group. Among them, model group and Astragalus flavonoids low, medium and high dose groups received reoxygenation for 24 hours after 2 hours of oxygen-glucose deprivation, and the low, medium and high dose groups were treated with different doses of Astragalus flavonoids at the same time of reoxygenation. The morphological changes of PC12 cells were observed by inverted phase contrast microscopy, and the survival rates of PC12 cells were measured by MTT and CCK-8 methods. Results: The cells in the normal group were in good condition, adherent growth and cell processes interlaced with each other were obvious, and the cells had strong refractive power and smooth surface. Compared with the normal group, some cells in the model group floated, the processes contracted, the refractive power of cells was poor, and the cell survival rates were significantly reduced ( P <0.05);compared with the model group, the cells in the Astragalus flavonoids low, medium and high dose groups were improved significantly with clear processes, and the cell survival rates were increased ( P <0.05).Compared with the low dose group, the medium and high dose groups had better cell status and higher cell survival rates ( P <0.05);there was no significant difference between the medium and the high dose group ( P > 0.05).Conclusion: Astragalus flavonoids can improve the growth status of OGD/R PC12 cells, alleviate cell damage, increase cell survival rate and play a protective ole, and the protective effect of Astragalus flavonoids medium and high dose groups is better than that in the low dose group.
