利用GEO在线数据构建脑胶质母细胞瘤ceRNA网络 被引量:2

摘要 目的 构建胶质母细胞瘤(GBM)ceRNA调控网络,探讨其发病机制。方法 利用GSE103229数据集的芯片数据,分析GBM中异常表达的lncRNA、mRNA和miRNA,差异基因判定标准为logFC>1,P<0.05。通过数据库分析三者之间的靶向调控关系,构建ceRNA网络并用Cytoscape软件可视化。结果 GBM差异基因中,lncRNA1326个,癌组织中上调743个,下调583个;差异mRNA3743个,癌组织中上调1865个,下调1878个;差异miRNA33个,上调9个,下调24个。构建了包含miR124、miR128、miR138和miR193b的GBM的ceRNA网络。结论 成功构建了GBM相关的ceRNA网络,其中miR124、miR128、miR138和miR193b是重要的节点基因。 Objective To construct a ceRNA regulatory network and explore its pathogenesis of glioblastoma (GBM). Methods Using the microchip data of GSE103229 dataset,we analyzed the abnormal expression arrays of lncRNA,RNA and microRNA in GBM.The screening criteria for differentially expressed genes were logFC>1 and P <0.05.Through the analysis of the targeted regulatory relationship between them,the ceRNA network was constructed and visualized by Cytoscape software. Results There were 1326 lncRNAs were differentially expressed,and 743 up regulated and 583 down regulated in GBM;3743 differentially expressed mRNAs,and 1865 up regulated and 1878 down regulated in GBM;33 differentially expressed miRNAs,and 9 up regulated and 24 down regulated.A ceRNA network containing miRNA 124,miRNA 128,miRNA 138 and miRNA 193b was constructed. Conclusion GBM related ceRNA networks have been successfully constructed,in which microRNA 124,microRNA 128,microRNA 138 and microRNA 193b are important node genes.
