摘要 目的本研究旨在探讨棕榈酸(PA)对H9C2心肌细胞凋亡的影响及活性氧水平的改变在其中的作用。方法不同浓度(0、50、100、200、400、800μmol/L)PA作用于心肌细胞18h,流式细胞术检测细胞早期凋亡率;DCFH-DA探针法检测细胞内活性氧水平。结果PA50μmol/L时细胞早期凋亡率有所增加;PA100μmol/L时,细胞早期凋亡率明显增加,活性氧水平明显增加;PA400μmol/L和800μmol/L时细胞密度出现大量减少。结论PA不同浓度作用于H9C2心肌细胞会导致细胞凋亡,其原因可能与PA增加细胞活性氧的产生有关。 Objective Although palmitic acid,the high concentration of saturated fatty acids in serum,has instigated the cardiovascular diseases,the underlying mechanisms remain unknown.The aim of this study was to elucidate that if palmitic acid induced the cardio-myocyte apoptosis and reactive oxygen species (ROS). Methods Different concentrations of PA(0,50,100,200,400 and 800μmol/L) were treated into myocardial cells for 18h,cell early apoptosis was detected by flow cytometry;ROS generation was measured by DCFH-DA assay. Results Treatment of palmitate acid in 50μmol/L elevated moderately cell apoptosis.Furtherly,Treatment of palmitate acid in 100μmol/L significantly increased cell apoptosis and ROS generation.Lastly,administration of palmitate acid in 400μmol/L and 800μmol/L decreased cell density,resulted in the decrease and even the none of cell apoptosis. Conclusion The intermediated concentration of palmitate acid induced cell apoptosis and the release of ROS,but high concentration induced cell death.
