摘要
采用索氏提取法从绿升麻中提取黄酮,利用有机溶剂进行萃取,通过AB-8型大孔吸附树脂进行纯化。利用高效液相色谱法测定含量,最佳色谱条件为:色谱柱ZORBAXEclipseXDB-C18色谱柱(250mm×4.6mmID,5μm)及保护柱(12.5mm×4.6mmID,5μm)流动相为色谱级乙腈和0.4%的磷酸水溶液,比例为13∶87;流速1mL/min;检测波长327nm;柱温25℃;室温;进样量10μL。结果:氯仿层在31.25~500g·mL^-1范围内;乙酸乙酯层在62.5~1000g·mL-1范围内;正丁醇层在31.25~500g·mL-1范围内呈现良好的线性关系。相对保留时间和相对保留峰面积的RSD值小于5%。分析可知,乙酸乙酯层中含有芦丁成分,氯仿层中含有槲皮素成分,正丁醇中含有芹菜素成分。
Flavonoids were extracted from Actaea asiatiea by soxhlet method,Petroleum were used for extraction,Ab-8 macroporous adsorption resin was used for purification,The content was determined by HPLC,The optimal chromatographic conditions were as follows: ZORBAX Eclipse XDB-C18 chromatographic column (250 mm×4.6 mm ID,5 μm) and protective column (12.5 mm×4.6 mm ID,5 μm). The mobile phase was chromatographically graded acetonitrile and 0.4% phosphoric acid aqueous solution,and the ratio was13 ∶ 87.The velocity of1 mL/min;Detection wavelength:327 nm;Column temperature25 ℃;At room temperature. Injection volume10 μL. Results:RSD values of relative retention time and relative retention peak area were less than 5%.Through the analysis of data results,the ethyl acetate layer contains rutin,the chloroform layer contains quercetin,and the n-butanol contains apigenin.
