微小RNA137基因与精神分裂症的关联研究 被引量:3

摘要 目的:探索微小RNA137(miR-137)基因与中国汉族精神分裂症及其临床表现之间的关系。方法:采用改良的多重连接酶检测反应技术(iMLDR)对254例稳定期精神分裂症患者(病例组)和339名健康对照者(对照组)miR-137基因rs1625579位点进行分型;采用阳性和阴性症状量表(PANSS)和重复性神经心理状态测验(RBANS)评定患者精神病性症状和认知功能,分析rs1625579位点与疾病及其症状间的关系。结果:病例组与对照组miR-137基因rs1625579位点基因型的观察值和期望值Hardy-Weinberg平衡吻合度良好(P=0.27;P=0.30);病例组和对照组比较miR-137基因rs1625579位点基因型和等位基因频率差异无统计学意义(χ~2=0.80,P=0.37;χ~2=0.75,P=0.39)。携带rs1625579位点G/T和T/T患者PANSS和RBANS评分比较差异无统计学意义(P均>0.05)。结论:本研究结果不支持miR-137基因是精神分裂症的易感基因。 Objective: To explore the relationship between microRNA137(miR-137) gene and schizophrenia and its clinical manifestations in Chinese Han population. Method:Improved Multiple Ligase Detection Reaction(iMLDR) technique was used to detect genotype polymorphisms of miR-137 rs1625579 in 254 patients(case group) with stable schizophrenia and 339 healthy subjects(control group);Positive and Negative Syndrome Scale(PANSS) and Repeatable Battery for the Assessment of Neuropsychological Status(RBANS) were used to evaluate the psychotic and cognitive symptoms in patients with schizophrenia and analyzed the relationship between rs1625579 and the disease and its symptoms. Results:The observed and expected values of miR-137 rs1625579 genotypes in the case and the control groups were in accordance with the Hardy-Weinberg equilibrium(P=0.27;P=0.30).There were no significant differences in miR-137 rs1625579 genotypes and the frequencies of allelic distribution between the case and the control groups(χ~2=0.80,P=0.37;χ~2=0.75,P=0.39).There were also no significant differences in PANSS and RBANS scores between patients with rs1625579 G/T and T/T(all P>0.05). Conclusion:The results of this study do not support miR-137 as a susceptibility gene for schizophrenia.
