摘要
目的检测甲基化转移酶(DNA methyltransferase,DNMT)DNMT1、DNMT2、DNMT3a、DNMT3bmRNA在乳腺癌组织及对应乳腺癌旁组织中的表达情况,分析其检测在乳腺癌发病机制及预后中的意义。方法应用实时荧光定量PCR(Flurescence quantitative-PCR,FQ-PCR)技术检测53例乳腺癌组织及20例癌旁组织中DNMT基因(DNMT1、DNMT2、DNMT3a、DNMT3b)mRNA的表达水平,并分析其表达与乳腺癌临床病理特征之间关系。结果乳腺癌组织中DNMT1、DNMT2、DNMT3a、DNMT3bmRNA的相对表达水平分别为0.57±0.27、0.58±0.29、0.56±0.27、0.61±0.26,而癌旁组织为0.20±0.10、0.17±0.12、0.18±0.13、0.19±0.17,癌组织均高于癌旁组织(t=8.10、7.57、5.34、7.92,P均<0.01),并且DNMT1、DNMT3a、DNMT3bmRNA的表达水平在淋巴转移是否之间的差异具有统计学意义(t=2.71、2.85、3.50,P均<0.05),而与乳腺癌其他临床病理特征参数之间差异均无有统计学意义(P均>0.05)。结论乳腺癌中DNMT基因表达的检测在可以为了解乳腺癌的发病机理提供参考依据,并且这种高表达的DNMT可能成为乳腺癌预后的指标之一。
Objective To detect the expression of methyltransferase DNMT1,DNMT2,DNMT3a and DNMT3b in breast cancer tissues and adjacent tissues, and to analyze the significance of detection in the pathogenesis and prognosis of breast cancer. Methods he expression levels of DNMT gene(DNMT1,DNMT2,DNMT3a, DNMT3b) in 53 breast cancer tissues and 20 adjacent breast cancer tissues were detected by real-time fluorescence quantitative PCR,and the relationship between the expression and clinicopathological characteristics of breast cancer was analyzed. Results The relative expression levels of DNMT1,DNMT2, DNMT3a and DNMT3b in cancer tissues were 0.57±0.27,0.58±0.29,0.56±0.27,0.61±0.26 and 0.20±0.10、0.17±0.12,0.18±0.13,0.19±0.17 in adjacent tissues respectively. The difference was statistically significant (t=8.10,7.57,5.34,7.92,all P<0.01). The expression of DNMT1、DNMT3a、DNMT3b mRNA were correlated with lymph node metastasis(t=2.71,2.85,3.50,P all <0.05) and were not correlated with other clinic pathologic parameters. Conclusion Detection of DNMT gene expression in breast cancer can provide a reference for understanding the pathogenesis of breast cancer, and this high expression of DNMT may become one of the prognostic indicators of breast cancer.
