欣胃颗粒及其拆方对胃癌前病变大鼠Axin、APC基因表达影响的研究

Effect of Xinwei granules and its disassembled prescriptions on Axin and APC gene expression in rats with precancerous lesion of gastric carcinoma

目的观察欣胃颗粒及其拆方对胃癌前病变模型大鼠胃黏膜中轴蛋白(Axin)、结肠腺瘤样息肉病基因(APC)表达的影响。方法选取360只Wistar健康雄性大鼠,按照随机数字表法将大鼠随机分为空白组、模型组、胃复春组和中药组,其中中药组又分为中药单法组、中药双法组、中药三法组和中药四法组。建立胃癌前病变大鼠模型,造模成功后给予各组大鼠相应处理,观察各组大鼠胃黏膜组织中Axin、APC基因的表达情况。结果与空白组比较,模型组、胃复春组和中药各组大鼠胃黏膜组织中AxinmRNA、APCmRNA的相对表达量均降低(P﹤0.05);与模型组比较,胃复春组和中药各组大鼠胃黏膜组织中AxinmRNA、APCmRNA的相对表达量均增高(P﹤0.05);与胃复春组比较,中药三法组和中药四法组大鼠胃黏膜组织中AxinmRNA的相对表达量均增高,中药双法组、中药三法组和中药四法组大鼠胃黏膜组织中APCmRNA的相对表达量均增高,差异均有统计学意义(P﹤0.05);与中药四法组比较,中药单法组、中药双法组、中药三法组大鼠胃黏膜组织中AxinmRNA、APCmRNA的相对表达量均降低(P﹤0.05)。结论欣胃颗粒可以上调胃癌前病变模型大鼠胃黏膜组织中Axin、APC基因的表达,其中中药四法组作用效果最强。

Objective To observe the effect of Xinwei granules and its disassembled prescriptions on the gene expression of Axin and adenomatous polyposis coli (APC) in rats with gastric precancerous lesions. Method Three hundred and sixty healthy male Wistar rats were included in the research and randomized as blank control group, model group, Weifuchun group and traditional Chinese Medicine (TCM) group, additionally, the TCM group was further subdivided as single TCM prescription group, two TCM prescriptions group, three TCM prescriptions group and four TCM prescriptions group, respectively. The gastric precancerous lesion models of rats were established and then treated correspondingly, and the expression of Axin and APC gene in rats’gastric mucosa tissue were observed. Result Compared with the blank control group, the expression levels of Axin mRNA and APC mRNA were significantly lower in model group, Weifuchun group and all TCM groups (P<0.05). Compared with the model group, the expression levels of Axin mRNA and APC mRNA were increased in Weifuchun group and all TCM groups (P<0.05);besides, the expression of Axin mRNA were significantly increased in three and four TCM prescriptions groups, and the APC mRNA were higher in two, three and four TCM prescriptions groups compared to Weifuchun group, with statistically significant differences observed (P<0.05);compared with four TCM prescriptions group, the expression of Axin mRNA and APC mRNA in gastric mucosa tissue of rats in single, two and three TCM prescriptions groups were decreased evidently (P<0.05). Conclusion Xinwei granules can up-regulate the expression levels of Axin and APC gene in rats with gastric precancerous lesions, and the four TCM prescriptions demonstrated the strongest effect.
