摘要
目的探讨乙二胺四乙酸二钾(EDTA-K2)抗凝管用于溶血标本胰岛素测定的价值。方法抽取糖尿病患者30例空腹静脉血各3管,使用普通凝胶干燥管制备血清为未溶血组,使用EDTA-K2抗凝管制备血浆为EDTA-K2抗凝组。使用普通凝胶干燥管制备Hb浓度为20g/L后,将Hb浓度调整为0.5、1、2.5、5、10g/L,分别加入未溶血组分为A1、B1、C1、D1和E1组,分别加入EDTA-K2抗凝组分为A2、B2、C2、D2和E2组。采用电化学发光法测定溶血处理后各组0.5、1、2、4 h胰岛素水平。结果未溶血组血清胰岛素水平与EDTA-K2抗凝组血浆胰岛素水平相仿(P>0.05)。A1组2、4 h和B1、C1、D1、E1组各时间点血清胰岛素水平均低于未溶血组(P<0.05),D2组和E2组各时间点血浆胰岛素水平低于EDTA-K2抗凝组(P<0.05)。结论 EDTA-K2抗凝管可以替代普通凝胶干燥管用于胰岛素水平检测,同时EDTA-K2抗凝血浆在标本发生Hb≤2.5 g/L溶血时可以稳定样本胰岛素水平,保证检测结果的准确性。
Objective To explore the value of anticoagulation tube with EDTA-K2 in detecting insulin in hemolysis samples.Methods Three tubes of fasting venous blood were taken from each of 30 patients with diabetes mellitus.Non-hemolysis group was prepared by normal dry tube and EDTA-K2-anticoagulated group was prepared by anticoagulation tube with EDTA-K2.Adding 0.5,1,2.5,5 and 10 g/L of Hb(erythroclasis happened in one tube to prepare 20 g/L of Hb) into non-hemolysis group to make groups of A1,B1,C1,D1 and E1,into EDTA-K2-anticoagulated group to make groups of A2,B2,C2,D2 and E2,orderly.Insulin level was detetected with electrochemiluminescence at 30 minutes,1st hour,2nd hour and 4th hour after hemolysis processing.Results Serum insulin level of non-hemolysis group was similar to plasma insulin level of EDTA-K2-anticoagulated group(P>0.05).Serum insulin levels of group A1 at the 2nd hour and 4th hour and the rest of groups at different time points were lower than those in non-hemolysis group(P<0.05).Plasma insulin levels of groups of D2 and E2 at different time points were lower than those in EDTA-K2-anticoagulated group(P<0.05).ConclusionThe EDTA-K2 anticoagulation tube can be used as an alternative to the normal dry tube for insulin detection.EDTA-K2 anticoagulated plasma can ensure the accuracy of the results by stabilizing insulin level of the samples with hemolysis of Hb≤2.5 g/L.
