摘要
目的探讨锌转运体LIV-1基因mRNA在肝癌组织中的表达及意义。方法对TCGA数据库中肝癌数据集的LIV-1基因的表达进行分析,采用逆转录PCR方法检测23例肝癌患者癌组织及其癌旁组织的LIV-1基因的mRNA表达水平进行验证。利用Kaplan Meier Plotter数据(http://kmplot.com/analysis/)中的肝癌数据集进行在线生存分析。通过生物信息学方法分析LIV-1基因启动子区的甲基化水平。结果TCGA数据库分析结果显示LIV-1 mRNA在肝癌组织中的表达显著高于正常肝脏组织(P<0.01),组织样本验证了生物信息学分析结果。相对于LIV-1低表达的肝癌患者,高表达LIV-1的肝癌患者的预后差(P<0.001)。相对于正常肝组织,LIV-1基因启动子区在肝癌组织中呈低甲基化(P<0.01)。结论异常高表达的LIV-1基因可能参与了肝癌的进展,是一种预后不良的因素。LIV-1基因启动子区低甲基化可能导致了其异常高表达。
Objective To explore the expression of LIV-1 mRNA and its significance in hepatocarcinoma.Methods The expression of LIV-1 in hepatocarcinoma data set of the TCGA database was analyzed.The mRNA level of LIV-1 in the paired tumor tissues and adjacent tissues of 23 patients with hepatocarcinoma was examined using reverse transcription PCR.Survival analysis in hepatocarcinoma data set was conducted by online analysis of Kaplan-Meier Plotter data(http://kmplot.com/analysis/).The methylation status in the promoter region of LIV-1 gene was bioinformatically analyzed.Results TCGA database analysis indicated that the expression of LIV-1 mRNA in hepatocarcinoma was significantly higher than that in normal liver tissues(P<0.01).Moreover,it was confirmed that LIV-1 was overexpressed in liver cancer tissues.The prognosis of hepatocarcinoma patients with higher LIV-1 mRNA level was much worse than that of patients with lower LIV-1 mRNA level(P<0.001).Compared to normal liver tissues,the promoter region of LIV-1 gene was hypomethylated in liver cancer(P<0.01).Conclusion The upregulated LIV-1 could promote the progression of hepatocarcinoma,and can be an indicator of poor prognosis.LIV-1 overexpression could result from the hypomethylation in the promoter region in hepatocarcinoma tissues.
