摘要
目的研究芝麻酚调控乳腺癌细胞MCF-7增殖的机理。方法采用不同浓度(0、20、50、100和250μmol·L^-1)芝麻酚处理乳腺癌细胞MCF-7,然后采用CCK8试剂盒检测细胞增殖情况;采用50μmol·L-1芝麻酚处理乳腺癌细胞MCF-748h,采用PI染色法,通过流式细胞仪分析细胞周期的变化,然后利用RT-qPCR对G1/S期转换的调控基因(p53、p21)进行检测。结果芝麻酚(50、100和250μmol·L^-1)能够显著抑制乳腺癌细胞MCF-7的增殖(P<0.05);芝麻酚处理组的S期细胞比例显著减少(由32.00%降到18.30%),G0/G1期细胞显著增加(由34.10%升到50.90%),以上结果表明发生G1/S期阻滞;RT-qPCR结果表明芝麻酚处理后,p53和p21的表达水平分别上调3.37和2.21倍(P<0.05)。结论芝麻酚显著抑制乳腺癌细胞MCF-7的增殖,芝麻酚可能通过上调p53和p21诱导G1/S期阻滞。
Objective To explore the mechanism of sesamol in regulating the proliferation of breast cancer cell MCF-7. Methods Breast cancer cells MCF-7 were treated with different concentrations (0,20,50,100 and 250 μmol·L^-1 ) of sesamol,and cell proliferation was detected by CCK8 kit.Breast cancer cells MCF-7 were treated with 50 μmol·L^-1 sesamol for 48 hrs.Cell cycle changes were analyzed by flow cytometry with PI staining and the expression of regulatory genes( p53、p21 ) in G 1/S transition phase was detected by RT-qPCR. Results Sesamol (50,100 and 250 μmol·L -1 ) can significantly ( P <0.05) inhibit the proliferation of breast cancer cells MCF-7;The proportion of S phase cells of treatment groups were significantly reduced (from 32.00% to 18.30 %),G 0/G 1 phase cells significantly increased (from 34.10% to 50.90%),and the above results showed that G1/S phase arrest occurred.The results of RT-qPCR showed that the expression levels of p53 and p21 were up-regulated by 3.37 and 2.21 times,respectively ( P <0.05). Conclusion Sesamol significantly inhibited the proliferation of breast cancer cells MCF-7 and induced G 1/S phase arrest possibly by up-regulating p53 and p21.
