摘要
灌胃方式下,研究富锌酵母提取物(ZnE)在大鼠体内的代谢动力学。大鼠分别单次、连续7d灌胃4mgZn/kg后,0.5、1、2、3、4、6、8、12、24、48、72、120h分别采集血液及心、肝、脾、肺、肾、胰、脑、睾丸、骨骼肌、股骨样品。利用高分辨电感耦合等离子体质谱法检测血浆中锌的含量,火焰原子吸收法测定组织中锌浓度,非房室模型计算ZnE的代谢动力学参数。ZnE在大鼠血液及体内各组织中末端消除半衰期大于238h,平均驻留时间大于59h,以股骨中锌浓度最高,其次是肝、胰、睾丸、肾、脾、肺、心、肌肉、脑。ZnE中的锌广泛分布在大鼠各组织中,消除缓慢。
Oral pharmacokinetics of zinc-enriched yeast extraction(ZnE) in rats was investigated. After ZnE was orally administered to rats at a single-dose and multiple-dose level of 4 mg Zn/kg, blood, heart, liver, spleen, lung, kidney, pancreas, brain, testes, muscle, and femur at 0.5, 1, 2, 3, 4, 6, 8, 12, 24, 48, 72, 120 h were collected for the quantification of zinc concentration. Noncompartmental analysis was employed to calculate the pharmacokinetic parameters after zinc concentrations in tissues were determined by flame atomic absorption spectrometry(FAAS) and plasma zinc levels were measured by high resolutioninductively coupled plasma-mass spectrometry(HR-ICP-MS). The terminal half-life(t1/2,λ) and mean residence time from 0 to time t(MRT0–t) of ZnE in the blood and the tissues were more than 238 h and 59 h, respectively. Zinc was widely distributed in various tissues with the level sequence of bone>liver>pancreas>testes>kidney>spleen>lung>heart>muscle>brain and eliminated slowly.
