补骨脂素对Aβ损伤PC12细胞的保护作用研究

Protective Effects of Psoralen on Aβ-induced PC12 Cells Injury

目的:研究补骨脂素对Aβ致PC12细胞损伤的保护作用及相关蛋白表达的影响,探讨其防治阿尔茨海默病的潜力。方法:将补骨脂素作用于Aβ损伤的PC12细胞,流式细胞仪检测细胞凋亡率,ELISA法检测Aβ含量,Western blot法检测β淀粉样前体蛋白(APP)、β分泌酶(BACE1)、雌激素受体β(ERβ)表达及p-ERK/ERK水平。结果:与模型组比较,补骨脂素组PC12细胞总凋亡率显著降低(P<0.01);ERβ蛋白表达量及p-ERK/ERK显著升高;Aβ1-42含量及APP、BACE1蛋白表达量显著降低(P<0.05或P<0.01)。结论:补骨脂素对Aβ损伤的PC12细胞具有保护作用,推测该作用机制为通过ERβ介导p-ERK信号通路进而使APP、BACE1及Aβ蛋白表达量降低。

Objective:To study the protective effects of psoralen on Aβ-induced PC12 cell injury and the expressions of related proteins,in order to explore its potential to prevent and treat Alzheimer′s disease.Methods:Psoralen was applied to Aβ-damaged PC12 cells,flow cytometry was used to detect apoptosis rate and ELISA was used to detect Aβ content,β amyloid precursor protein(APP),β secretase(BACE1),estrogen receptor β(ERβ)expressions and p-ERK/ERK level were detected by Western blot.Results:Compared with the model group,the total apoptosis rate of PC12 cells in psoralen group was significantly decreased(P<0.01),the expression of ERβ protein and p-ERK/ERK were significantly increased,the content of Aβ1-42 and APP,BACE1 proteins expressions decreased significantly(P<0.05 or P<0.01).Conclusion:Psoralen has protective effect on PC12 cells injured by Aβ,it is speculated that this mechanisum is to reduce the expressions of APP,BACE1 and Aβ proteins through ERβ-mediated p-ERK signaling pathway.
