HPLC测定不同产地昆明山海棠中的雷公藤红素 被引量:4

Determination of tripterine in Tripterygium hypoglaucurn from several habitats by HPLC

摘要 目的采用HPLC法测定多个产地野生昆明山海棠Tripterygium hypoglaucum(Levl.)Hutch中雷公藤红素的含量,并进行含量比较。方法采用Chromplus■C18色谱柱(250 nm×4.6μm,5μm),流动相为乙腈-0.1%磷酸水溶液(80∶20),流速1.0 mL·min^-1,检测波长425 nm,柱温25℃,进样量20μL。结果2×10^-5~0.5 mg·mL^-1雷公藤红素的线性关系良好(r=0.9990),平均回收率为87.00%,RSD=1.52%(n=6)。7个产地21批昆明山海棠中雷公藤红素含量最高的为0.82‰,最低的为0.12‰,含量的平均值为0.33‰。结论所用方法操作简便、分析快速、重复性好、准确度高,可作为昆明山海棠药材中雷公藤红素的定量分析方法,为该类药材的合理运用提供一定依据;建议设定昆明山海棠中雷公藤红素的含量不低0.1‰。 OBJECTIVE To determine tripterine in Tripterygium hypoglaucum(Levl.)Hutch from several habitats by HPLC,and to compare their tripterine contents.METHODS The high performance liquid chromatography external standard method was adopted with chromatographic column of chromplus■C18 column(250 nm×4.6μm,5μm).Mobile phase consisted of 0.1%phosphoric acid water solution(80∶20)with a flow rate of 1.0mL·min^-1,detection wavelength of 425 nm,column temperature of 25℃and sampling amount of 20μL.RESULTS The linear relationship was good(r=0.9990)in the range of 2×10^-5 to 0.5 mg·mL^-1 with the average recovery of 87%and the RSD of 1.52%(n=6).The highest content of tripterine in the 21 batches of T.hypoglaucum seven origins was 0.82‰,while the lowest content was 0.12‰and the overall average content of tripterine was 0.33‰.CONCLUSION The HPLC method is simple in operation,fast in analysis,good in reproducibility and high in accuracy.It can be used as a quantitative method for the quantitative analysis of tripterine in T.hypoglaucum,and it provided a basis for the rational use of wild T.hypoglaucum,and it is recommended that the content of tripterine of this kind of traditional Chinese medicine should not be lower than 0.1‰.
