摘要
为探讨水貂毛色差异形成的分子机制,试验利用美洲水貂肌肉组织对其黑色素皮质激素受体-1(MC1R)基因CDS区进行PCR扩增,并利用DNAMAN、ProtParam、TMHMM等生物信息学软件分析其氨基酸组成、与其他物种的同源性、跨膜区段等。结果表明:克隆获得的MC1R序列长度为954bp,编码317个氨基酸;美洲水貂MC1R基因序列与欧洲水貂的同源性为95.49%,美洲水貂MC1R基因编码的氨基酸序列与欧洲水貂同源性为92.43%;得到带负电氨基酸13个,带正电氨基酸22个,极性氨基酸67个,疏水性氨基酸157个,为亲水性蛋白质;共得到7个蛋白质的跨膜结构域。
In order to determine the molecular mechanism of mink hair color difference, PCR amplification was carried out in the CDS region of the melanin corticosteroid receptor 1 ( MC1R) gene of Neovisonvison by using muscle tissue of mink. It also used bioinformatics software such as DNAMAN, ProtParam, and TMHMM to analyze its amino acid composition, homology with other species, and transmembrane segments.The results showed that the MC1R CDS sequence is 954 bp long and encodes 317 amino acids.The homology of MC1R gene sequence with European mink is 95. 49%,and that of amino acid sequence encoded by MC1R gene with European mink is 92. 43%. There are 13 negatively charged amino acids, 22 positively charged amino acids, 67 polar amino acids and 157 hydrophobic amino acids, which are hydrophilic proteins. A total of 7 protein transmembrane domains are obtained.
