摘要
目的调查福建省不同地域白纹伊蚊线粒体细胞色素C氧化酶亚基I(mtDNA-CO1)基因的遗传多态性和系统发育关系。方法2015-2017年采集福建省15个媒介监测点的白纹伊蚊成蚊和幼虫样品,提取基因组DNA,扩增全长mtDNA-CO1基因片段并测序,应用Mega6.0和DnaSP5.0软件做基因特征分析和系统发育分析。结果新得到54份白纹伊蚊mtDNA-CO1基因片段序列,用于分析的片段长度为1388 bp。样本间多序列比对显示8个核苷酸差异位点,单倍型多样性为0.746,核苷酸多样性为0.126%。根据单倍型参考序列构建系统发育树,确定福建省白纹伊蚊存在9种单倍型,即:H01、H02、H03、H04、H08、H09、H12、H17、H23,另有6条序列无法确定单倍型。结论根据mtDNA-CO1基因多样性,2015-2017年福建省媒介监测点采集白纹伊蚊分属9种单倍型,这对于当地白纹伊蚊的种群进化研究及相关虫媒病毒病防控具有重要参考价值。
Objective To investigate the genetic diversity and phylogenetic relationships of the mitochondrial cytochrome c oxidase subunit I(mtDNA-CO1)gene in Aedes albopictus from Fujian province.Methods Mosquito samples from fifteen vector surveillance sites were collected in Fujian during 2015-2017,full-length mtDNA-CO1 genes were amplified and sequenced,Mega6.0 and DnaSP5.0 were used for homology analysis.Results Multiple sequence alignment indicated 8 variations at the nucleotide level among a total of 54 newly obtained fragments and another 12 amplicons from dengue surveillance sites in 2016.Haplotype diversity(Hd=0.746)and nucleotide diversity(π=0.126%)were determined.Based on phylogenetic analysis with 72 reference sequences of known haplotypes,nine haplotypes including H01,H02,H03,H04,H08,H09,H12,H17 and H23 were identified in 60 Ae.albopictus samples,while another six samples could not be assigned to known haplotypes.Conclusion These data demonstrated genetic diversity in the mtDNA-CO1 genes of Ae.albopictus collected from vector surveillance sites in Fujian province.These results facilitate research on population evolution of Ae.albopictus,as well as prevention and control of related arboviral diseases.
