补肾养血方防治气态甲醛慢性染毒小鼠卵巢损伤的研究 被引量:4

Study of Bushen Yangxue Recipe (补肾养血方) on Prevention and Treatment of Ovarian Injury in Mice by Chronic Poisoning with Gaseous Formaldehyde

摘要 目的:研究气态甲醛慢性染毒对小鼠卵巢功能的影响,以及补肾养血方对其卵巢毒性的防治作用。方法:将30只性成熟SPF级雌性昆明小鼠随机分为对照组、染毒组和治疗组,对照组给予吸入经过滤的新鲜空气,染毒组和治疗组采用气态甲醛动态吸入式染毒,6 h/d,连续14 d,同时治疗组给予补肾养血方灌胃,对照组和染毒组给予等量生理盐水灌胃。观察小鼠一般状态及动情周期。实验结束后处死小鼠,测定小鼠血清性激素水平,计算卵巢脏器系数,观察卵巢病理形态学变化,并计数各级卵泡数,检测卵巢局部相关基因的表达水平。结果:染毒组小鼠一般状态差,动情周期延长,小鼠血清E2水平降低,FSH、LH水平升高,卵巢脏器系数降低,卵巢内生长卵泡减少,闭锁卵泡增多;卵巢Foxl2、FSHR mRNA表达水平升高,Esr2、Pgr mRNA表达水平降低,与对照组比较,差异有统计学意义(P<0.05)。治疗组小鼠一般状态好转,动情周期缩短;血清E2水平升高,FSH水平降低,卵巢脏器系数升高,卵巢内生长卵泡增加,闭锁卵泡减少,卵巢Foxl2、FSHR mRNA表达降低,Esr2 mRNA表达上升,与染毒组比较,差异有统计学意义(P<0.05)。Pgr mRNA表达有上升趋势,但与染毒组比较差异无统计学意义(P>0.0)。结论:气态甲醛慢性染毒可影响小鼠卵巢功能,补肾养血方可防治甲醛对卵巢的损害作用,改善卵巢功能。 Objective:To study the effect of chronic exposure to gaseous formaldehyde on ovarian function in mice and the preventive and therapeutic effects of Bushen Yangxue recipe on its ovarian toxicity.Methods:A total of 30 sexually mature SPF female Kunming mice were randomly divided into control group,exposure group and treatment group.The control group received inhaled filtered fresh air.The exposed group and the treatment group were treated with gaseous formaldehyde dynamic inhalation.The gaseous formaldehyde was administered for 6 h/d for 14 days.At the same time,the treatment group was given Bushen Yangxue recipe,and the control group and the exposed group were given the same amount of normal saline.Observed the general state and the estrous cycle of the mice.At the end of the experiment,the mice were sacrificed,serum sex hormone levels were determined,ovarian organ coefficients were calculated,pathological changes of ovary were observed,and the number of follicles at various levels were counted to detect the expression levels of ovarian local related genes.Results:The mice in the exposed group were generally in poor condition,and the estrous cycle was prolonged.The levels of serum E2 were decreased,the levels of FSH and LH were increased,the ovarian organ coefficient were decreased,the follicles in the ovary were decreased,and the atresia follicles were increased.The expression levels of Foxl2 and FSHR mRNA in the ovarian were increased,and the levels of Esr2 and Pgr mRNA were decreased,and the difference was statistically significant(P<0.05).In the treatment group,the general condition of the mice were improved,the estrous cycle were shortened;the serum E2 levels were increased,the FSH levels were decreased,the ovarian organ coefficient were increased,the ovarian growth follicles were increased,the atresia follicles were decreased,the ovary Foxl2,FSHR mRNA expression were decreased,and the levels of Esr2 were increased.There was a statistically significant difference between the two groups(P<0.05).The expression of Pgr mRNA was increased,but there was no significant difference between the two groups(P>0.05).Conclusion:Chronic exposure to gaseous formaldehyde can affect the ovarian function of mice.Bushen Yangxue recipe can prevent the damage of formaldehyde on the ovary and improve ovarian function.
