大连海域海洋功能细菌的筛选

Screening of Marine Functional Bacteria in Dalian Sea Area

摘要 从大连海域采集30余种海洋样品,用海水2216E固体培养基、含亚甲基蓝的固体培养基、含碳酸钙的固体培养基等进行培养,初筛出100多株具有产酸、产酶、产抗氧化性物质的菌株;20℃培养48 h,复筛出12株繁殖力强的低温菌,对其进行生长量检测、革兰氏染色、产酸、产葡萄糖氧化酶、亚硝酸盐降解、硝酸盐分解及10℃低温培养。其中菌株M2,5,6,4-1,7-1耐低温、生长快、产酸高、降解亚硝酸能力强、不分解硝酸盐、革兰氏阳性,兼性厌氧,初步确定为低温乳酸菌。同时还筛选出40余株具有葡萄糖氧化酶特性的菌株以及其他特性菌株,并对筛选的近200株海洋菌株进行保藏。该研究进一步证明了海洋微生物种类的多样性,使人们对未知的海洋微生物世界的研究更加着迷。 In this paper, more than 30 marine samples were collected from Dalian sea area and cultured using 2216E solid medium, methylene blue solid medium and calcium carbonate solid medium. More than 100 bacterial strains with acid, enzyme and antioxidant properties were screened out. After 48 hours of incubation at 20 °C, 12 bacterial strains of low temperature with strong fecundity were screened out, and their growth rate test, Gram staining, acid production, glucose oxidase production, nitrite degradation, nitrate decomposition and low temperature culture at 10 °C were performed. Among them, strains M2, 5, 6, 4-1, 7-1 are cold-resistant, fast-growing, acid-producing, strong ability to degrade nitrite, non-decomposition nitrate, Gram-positive, facultative anaerobic, and are initially identified as low-temperature lactic acid bacteria. At the same time, more than 40 strains with the characteristics of glucose oxidase and other strains were screened, and nearly 200 marine strains were screened for preservation. This study further proves the diversity of marine microbial species and makes people more fascinated to the unknown marine microbial world.
