电针预处理对急性心肌梗死再灌注无复流大鼠黏附分子及内皮细胞功能的影响 被引量:5

Effect of electroacupuncture preconditioning on adhesion molecules and endothelial cells function in rats with acute myocardial infarction with reperfusion and no reflow

摘要 目的观察电针预处理对急性心肌梗死再灌注无复流大鼠细胞黏附分子及内皮细胞功能的影响。方法将40只雄性SD大鼠随机分为假手术组、模型组、电针预处理组、替罗非班组,除假手术组外,其余组大鼠均采用结扎左冠状动脉45min后再灌注120min的方法复制急性心肌缺血再灌注无复流模型。实验结束后观察各组大鼠心肌梗死及无复流的范围,采用酶联免疫吸附法检测心肌组织中肌钙蛋白I(cTnI)、肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)水平,并通过蛋白质印迹法检测心肌组织中黏附分子[整合素β1(Integrinβ1)、P选择素(P-selectin)、E选择素(E-selectin)]和内皮细胞功能[血管内皮生长因子(VEGF)、内皮型一氧化氮合酶(eNOS)、一氧化氮(NO)]相关蛋白表达水平。结果电针预处理组心肌无复流率和梗死率均明显低于模型组(P均<0.05);心肌组织中cTnI、CK、CK-MB水平和VEGF、eNOS、NO蛋白表达水平均明显低于模型组(P均<0.05),Integrinβ1、E-selectin和P-selectin蛋白表达水平均明显高于模型组(P均<0.05)。结论电针内关、心俞、膻中能够通过调控心肌组织中细胞黏附分子和内皮细胞功能因子的表达,从而减少再灌注无复流现象,缩小心肌梗死面积,起到保护心脏的作用。 Objective It is to observe the effects of electroacupuncture preconditioning on cell adhesion molecules and endothelial cell function in rats with acute myocardial infarction with reperfusion and no reflow. Methods Forty male SD rats were randomly divided into sham operation group, model group, electroacupuncture pretreatment group and tirofiban group. Except for sham operation group, the other groups were treated with ligation of left coronary artery for 45 min and then reperfusion for 120min method to establish the no-reflow model of acute myocardial ischemia-reperfusion. At the end of the experiment, the range of myocardial infarction and no-reflow in each group was observed. The troponin I (cTnI), creatine kinase (CK) and creatine kinase isoenzyme (CK-MB) were detected by enzyme-linked immunosorbent assay, the expression levels of related protein such as adhesion molecules in myocardial tissue [Integrinβ1, P-selectin, E-selectin] and endothelial cell function [vascular endothelial growth factor (VEGF), endothelial nitric oxide synthase (eNOS), nitric oxide (NO)] were detected by western blotting methods. Results The myocardial no-reflow rate and infarction rate, the levels of cTnI, CK, CK-MB and VEGF, eNOS and NO in myocardial tissue were significantly lower in the electroacupuncture preconditioning group than that in the model group ( P <0.05), the expression levels of Integrin β 1, E-selectin and P-selectin were significantly higher than those in the model group ( P <0.05). Conclusion Electroacupuncture on Neiguan, Xinshu and Tanzhong can regulate the expression of cell adhesion molecules and endothelial cell functional factors in myocardial tissue,thus to reduce the phenomenon of reperfusion without reflow, reduce myocardial infarct area and protect the heart.
