复方芩兰口服液儿童用药最佳有效剂量的研究

Optimal Effective Dose of Coumpound Qinlan Oral Liquid in Children

目的:探讨复方芩兰口服液儿童用药最佳有效剂量,为儿童临床应用提供参考。方法:采用已知成人用量换算儿童用量的计算方法,设定1~12岁儿童的用药范围,通过甲型H1N1流感病毒(FM1株)滴鼻感染致幼龄小鼠肺炎模型及死亡模型观察其抗病毒作用;采用腹腔毛细血管通透性实验观察其抗炎作用;采用冰醋酸致幼龄小鼠扭体实验观察其镇痛作用;采用氨水致幼龄小鼠咳嗽模型观察其止咳作用,从而对已设定剂量范围的有效性进行评价。结果:复方芩兰口服液0.2、0.5、0.7、0.9、1.1、1.4 mL·(kg·d)^-1 6个剂量组均可降低感染所致幼龄小鼠死亡率、延长平均存活天数,减少醋酸致幼龄小鼠疼痛的扭体次数,与模型对照组比较有显著差异(P<0.05、P<0.01)。0.2、0.5、0.7、0.9 mL·(kg·d)^-1 4个剂量组可明显降低肺指数,与模型对照组比较有显著性差异(P<0.05)。0.5、0.7、0.9、1.1、1.4 mL·(kg·d)^-1 5个剂量组可明显抑制冰醋酸引起的幼龄小鼠腹腔毛细血管通透性增高,与模型对照组比较有显著差异(P<0.01)。结论:复方芩兰口服液在0.5~1.4 mL·(kg·d)^-1剂量范围内对幼龄小鼠模型均具有良好的抗病毒、抗炎、镇痛、止咳的作用。

Objective: To explore the optimal effective dose Compound Qinlan Oral Liquid in children, thus to provide its clinical reference in pediatrics. Methods: Set the dosage range for 1~12 years old children according to adult dosage, its anti-viral effect was observed by the pneumonic model and mortality model;its anti-inflammatory effect was observed by abdominal capillary permeability model;its analgesic effect was observed by glacial acetic acid-writhing tests, and its antitussive effect was observed by ammonia water steaming test. The effectiveness of the set dosage was evaluated based on the above tests. Results: All the six concentrations(0.2, 0.5, 0.7, 0.9, 1.1, 1.4 mL/kg·d) significantly decreased the death rate and prolonged the life span of young mice induced by FM1, as well as greatly reduced writhing times of young mice induced by glacial acetic acid, there were statistical difference compared to those in the model group( P <0.05, P <0.01). Four concentrations(0.2, 0.5, 0.7, 0.9 mL/kg·d) could significantly decrease lung index compared to the model group( P <0.05). Five concentrations(0.5, 0.7, 0.9, 1.1, 1.4 mL/kg·d) showed obvious inhibition effect on capillary permeability increase compared to the model group( P <0.01). Conclusion: Compound Qinlan Oral Liquid with the concentrations of 0.5~1.4 mL·(kg·d)^-1 have better antiviral, antiinflammatory, analgesic and antitussive effects.
