虎杖提取物血脂调节作用及其机制研究 被引量:17

摘要 目的:研究虎杖苷的血脂调节作用,并探讨其可能机制。方法:SD大鼠建立高脂血症模型,灌胃给予虎杖苷100 mg/kg,1次/d,连续给药4周。测定大鼠TC、TG、LDL-C、HDL-C、ox-LDL、NO、MDA浓度及CAT、SOD活性,测定CPT-I、CPTII、ACC和FAS水平;观察肝脏组织学形态变化。结果:与空白对照组相比,模型对照组TC、TG、LDL-C、ox-LDL、MDA、CPI-I、CPI-II、FAS和ACC水平显著升高(P<0.05),HDL-C、NO、HMGR水平显著降低(P<0.05),CAT、SDO活性显著降低(P<0.05),肝脏脂质堆积明显。虎杖苷组TC、TG、LDL-C、ox-LDL、HDL-C水平较模型对照组显著改善(P<0.05),与阳性对照组无显著性差异(P>0.05);虎杖苷组CAT、SOD活性显著升高(P<0.05),MDA活性显著降低(P<0.05),NO、HMGR水平显著升高(P<0.05),CPI-I、CPI-II、FAS和ACC表达均显著降低(P<0.05);虎杖苷组adipo R2和PPARαmRNA表达水平提高(P<0.05);虎杖苷组大鼠肝脏脂肪变性程度病变最轻。结论:虎杖苷可以有效降低高脂血症大鼠的血脂水平,改善肝脏脂肪病变,其机制可能与减少脂肪酸和胆固醇合成,纠正高脂血症大鼠氧化应激状态,从而减少脂质过氧化,防止脂肪堆积。 Objective:To study the blood lipids regulation of polydatin and to explore its possible action mechanism.Methods:48 SD rats were randomly divided into the blank control group,the model group,the positive control group and the polydatin group,with 12 rats in each group.The rats in the blank control group were fed normally,and the other groups were fed with high fat diet.The rat model of hyperlipidemia was established 4 weeks later.From the 5th week,the rats in the positive control group were given simvastatin 6 mg/(kg·d),and the rats in the polydatin group were intragastrically administered with 100 mg/kg of polydatin.Rats in the blank control group and the model group were given distilled water once a day for 4 weeks.The concentrations of TC,TG,LDL-C,HDL-C,ox-LDL,NO,MDA,and the activity of CAT and SOD were measured;also the levels of CPT-I,CPT-II,ACC and FAS were determined.Histological changes of liver were observed in the rats by HE staining.Results:Compared to the blank control group,the levels of TC,TG,LDL-C,ox-LDL,MDA,CPI-I,CPI-II,FAS and ACC were significantly increased in the model control group(P<0.05);the levels of HDL-C,NO,and HMGR were significantly decreased(P<0.05);the activities of CAT and SDO were also significantly decreased(P<0.05).Liver lipid accumulation was obvious.The levels of TC,TG,LDL-C,ox-LDL and HDL-C in the polydatin group were significantly improved compared to those in the model group(P<0.05);of which there were no statistical differences between the positive control group and the polydatin group(P>0.05).In the polydatin group,the activities of CAT and SOD were significantly increased(P<0.05),the activity of MDA was significantly decreased(P<0.05);the levels of NO and HMGR were significantly increased(P<0.05);the expressions of CPI-I,CPI-II,FAS and ACC were significantly decreased(P<0.05);the mRNA levels of adipo R2 and PPARαwere significantly increased(P<0.05);the degree of liver steatosis was the lightest.Conclusion:Polydatin can effectively reduce the blood lipids level and improve liver fat lesions in hyperlipidemic rats.The mechanism may be related to reducing fatty acid and cholesterol synthesis,correcting the oxidative stress state of hyperlipidemia rats,and thus to reduce lipid peroxidation and prevent fat accumulation.
